摘要
目的建立HPLC法同时测定新会柑Citrus reticulata‘Chachi’种子中辛弗林、没食子酸、橙皮苷、柠檬苦素、川陈皮素、3,5,6,7,8,3′,4′-七甲氧基黄酮、橘皮素的含有量。方法新会柑种子甲醇提取物的分析采用DIKMA Diamonsil C18柱(250 mm×4.6 mm,5μm);流动相磷酸水-(甲醇∶乙腈=1∶1),梯度洗脱;体积流量1.0 mL/min;柱温30℃;检测波长辛弗林(224 nm),没食子酸(218 nm),橙皮苷(283 nm),柠檬苦素(209 nm),川陈皮素、3,5,6,7,8,3′,4′-七甲氧基黄酮、橘皮素(330 nm)。结果 7种成分在各自范围内线性关系良好(R2≥0.999 1),平均加样回收率98.546%~101.769%, RSD 1.3%~2.6%。结论该方法准确稳定,重复性好,可用于新会柑种子的质量控制。
AIM To establish an HPLC method for the simultaneous content determination of synephrine, gallic acid,hesperidin,limonin,nobiletin,3,5,6,7,8,3',4'-heptamethoxy flavonoids,and tangeretin in the seeds of Citrus reticulata‘Chachi’. METHODS The analysis of methanol extract of the seeds from C. reticu-lata was performed on a 30 ℃ thermostatic DIKMA Diamonsil C18 column ( 250 mm×4. 6 mm,5μm),with the mobile phase comprising of phosphoric acid-( methyl alcohol ∶ acetonitrile = 1 ∶ 1) flowing at 1. 0 mL /min in a gradient elution manner,and the detection wavelengths were set at synephrine ( 224 nm),gallic acid ( 218 nm), hesperidin ( 283 nm),limonin ( 209 nm),nobiletin,3,5,6,7,8,3',4'-heptamethoxy flavonoids and tan- geretin ( 330 nm). RESULTS Seven constituents showed good linear relationships within their own ranges ( R2≥ 0. 999 1 ),whose average recoveries were 98. 546%- 101. 769%,with the RSDs of 1. 3%- 2. 6%. CONCLUSION This accurate,stable and reproducible method can be used for the quality control of the seeds from C. reticulata.
