小米多酚对α-淀粉酶和α-葡萄糖苷酶的体外抑制活性及降糖潜力研究

摘要 为探究小米多酚的降糖效果,以α-淀粉酶和α-葡萄糖苷酶抑制率为指标,探究其对2种酶抑制活性的稳定性,并以高糖诱导的2型糖尿病秀丽隐杆线虫为模型测定体内甘油三酯和葡萄糖含量。结果表明:小米多酚提取物在体外对α-淀粉酶和α-葡萄糖苷酶都具有一定的抑制效果,其抑制作用与质量浓度呈明显的剂量-效应关系,抑制类型均为可逆性混合型抑制作用,并在30~40℃、pH 3~5条件下最有利于发挥抑制活性。小米多酚干预显著降低了高糖诱导的线虫体内的甘油三酯和葡萄糖含量。总体表明,小米多酚具有潜在的降血糖作用。 To investigate the hypoglycemic effect of millet polyphenols,the inhibition rates ofα-amylase andα-glucosidase were used as indicators to evaluate the stability of their inhibitory activity on these two enzymes,and the triglyceride and glucose contents were measured in a high-glucose-induced type 2 diabetic Caenorhabditis elegans model.The results showed that the millet polyphenol extract exhibited certain inhibitory effects on bothα-amylase andα-glucosidase in vitro.The inhibition was found to demonstrate a clear dose-effect relationship with mass concentration,with both inhibitions identified as reversible mixed-type inhibition.The optimal conditions for inhibitory activity were determined to be 30~40℃and pH 3~5.Intervention with millet polyphenols significantly reduced the levels of triglycerides and glucose in Caenorhabditis elegans induced by high glucose.It was concluded that millet polyphenols possess potential hypoglycemic effects.
