摘要
采用气相色谱-串联三重四极杆质谱(GC-MS/MS)法测定化妆品中乳酸、乙醇酸、2-羟基丁酸、甘油酸、羟基辛酸、苹果酸、扁桃酸、酒石酸、柠檬酸、二苯乙醇酸、葡萄醛酸11种α-羟基酸含量。用N,N-二甲基甲酰胺提取化妆品中α-羟基酸,经N,O-双-(三甲基硅烷基)三氟乙酰胺(BSTFA)硅烷化衍生后,用VF-1701ms毛细管柱(30 m×0.25mm×0.25μm)分离,采用GC-MS/MS法在多反应监测模式下检测,用外标法定量。11种α-羟基酸的质量浓度在0.5~5μg/mL范围内与定量离子色谱峰面积线性关系良好,相关系数均不小于0.995 5,方法检测限和定量限分别为0.05~0.2μg/mL和0.2~0.6μg/mL。4种不同基质(液体类、膏霜类、乳液类、贴膜类)样品在添加质量浓度分别为1、1.5、2.5μg/mL时的平均回收率为76.2%~125%,测定结果的相对标准偏差均不大于14%(n=6)。该法可用于化妆品中11种α-羟基酸的含量测定。
A method of gas chromatography-tandem triple quadrupole mass spectrometry(GC-MS/MS)was establish ed for the determination of 11α-hydroxy acids,including lactic acid,glycolic acid,2-hydroxybutyric acid,glyceric acid,hydroxyoctanoic acid,malic acid,mandelic acid,tartaric acid,citric acid,benzilic acid and glucuronic acid,in cosmetics.Theα-hydroxy acids in cosmetics were extracted with N,N-dimethylformamide,and after silylation derivatization with N,O-bis-(trimethylsilyl)trifluoroacetamide(BSTFA),theα-hydroxy acids were separated using a VF-1701ms(30 m×0.25 mm×0.25μm),and detected by GC-MS/MS in multi-reaction monitoring mode,and quantified by the external standard method.The mass concentrations of 11α-hydroxy acids had good linear relationship with the peak areas of the quantitative ions in the range of 0.5-5μg/mL.The correlation coefficients were not less than 0.9955.The detection limits and quantification limits of the method were 0.05-0.2μg/mL and 0.2-0.6μg/mL,respectively.The average recoveries of four different matrices(liquid,cream,lotion and film)at spiked levels of 1,1.5,2.5μg/mL were 76.2%-125%,and the relative standard deviations of the determination results for six parallel samples were not exceeding 14%.The method can be used for the determination of 11α-hydroxy acids in cosmetics.
