摘要
目的分析不同表型艾叶内生菌群落、关键黄酮类和酚酸类成分的特征,通过高丰度群落与关键成分的相关性分析,从微生物组的角度阐释艾叶内生菌对其药材品质的影响。方法运用高通量扩增子技术分析不同表型艾叶内生菌群落组成和多样性;利用高效液相色谱技术和酶标法检测不同表型艾叶关键成分的含量;利用斯皮尔曼相关性分析筛选艾叶关键成分显著相关的内生菌群落。结果不同表型的艾叶内生细菌和真菌的群落多样性呈现显著性差异;通过LEfSe分析,确定鞘脂菌属(Sphingobium)、假单胞菌属(Pseudomonas)、马赛菌属(Massilia)、马氏黑粉菌属(Macalpinomyces)、枝孢属(Cladosporium)等为不同表型艾叶生物标识类群。相关性分析表明,马赛菌属(Massilia)、泛菌属(Pantoea)、链格孢属(Alternaria)、枝孢属(Cladosporium)、镰刀菌属(Fusarium)等属的丰度与部分关键黄酮类和酚酸类成分的含量显著性正相关(P<0.001)。结论本研究阐明了不同表型艾叶内生菌及关键成分的分布特征,并初步确定了与其关键成分显著正相关的群落,为进一步分离促进艾叶关键成分积累的内生菌株,提升艾叶的品质提供了科学依据。
Objective To investigate the characteristics in endophytic communities and key flavonoid and phenolic acid components among different phenotypes of Artemisia argyi leaves,the impact of endophytes on the quality of A.argyi leaves was analyzed from the perspective of microbiome through correlation analysis.Methods Amplicon sequencing was utilized to analyze the diversity and composition of the A.argyi leaves with different phenotypes.The key metabolites of A.argyi leaves with different phenotypes were detected by high-performance liquid chromatography and ultraviolet spectrophotometry.To identify endophytic communities strongly associated with key metabolites,Spearman’s correlation analysis was subsequently employed.Results There were significant differences in the diversity and composition of endophytic bacteria and fungi in A.argyi leaves with different phenotypes.According to linear discriminant analysis effect size(LEfSe)analysis,Sphingobium,Pseudomonas,Massilia,Macalpinomyces,and Cladosporium were identified as potential microbial markers.Correlation analysis revealed that the abundances of Massilia,Exiguobacterium,Pantoea,Alternaria,Cladosporium,and Fusarium were significantly and positively correlated with the contents of some key flavonoids and phenolic acids(P<0.001).Conclusion The findings delineated that the distribution characteristics of endophytic communities and key metabolites of A.argyi leaves with different phenotypes.Several endophytic communities were identified to be strongly associated with key metabolites,which facilitates future isolation of these functional strains and potential quality optimization of A.argyi leaves.
