中医药调控腺苷酸活化蛋白激酶信号通路防治阿尔茨海默病研究进展

Prevention and Treatment of Alzheimer′s Disease with Traditional Chinese Medicine through Adenosine Monophosphate-activated Protein Kinase Signaling Pathway:A Review

摘要阿尔茨海默病(AD)是临床常见的神经退行性疾病,严重威胁患者的健康和生命质量,目前临床尚无根治手段,亟须寻求治疗AD的有效药物。腺苷酸活化蛋白激酶(AMPK)参与调节机体的氧化应激、脂代谢、炎症、自噬及线粒体功能障碍等进程,与AD发生发展关系密切。中医药在防治AD方面具有独特优势。大量研究表明,中医药通过调控AMPK信号通路从而发挥恢复线粒体功能、调节脂代谢、抑制炎症、调节自噬、减轻氧化应激等多重作用,达到治疗AD的目的。本文通过检测近年来中医药治疗AD的相关文献,总结了AMPK结构及其影响AD的机制,并对中医药调控AMPK信号通路防治AD的研究现状进行综述,以期为AD的防治提供理论基础和新思路。 Alzheimer′s disease(AD)is a common neurodegenerative disorder that severely threatens patients′health and quality of life.Currently,there is no curative treatment,making it urgent to identify effective drugs for AD therapy.Adenosine monophosphate-activated protein kinase(AMPK)participates in regulating oxidative stress,lipid metabolism,inflammation,autophagy,and mitochondrial dysfunction,and is closely associated with the occurrence and progression of AD.Traditional Chinese medicine(TCM)has unique advantages in preventing and treating AD.Numerous studies have shown that TCM can modulate the AMPK signaling pathway to restore mitochondrial function,regulate lipid metabolism,inhibit inflammation,modulate autophagy,and reduce oxidative stress,thereby achieving therapeutic effects in AD.This paper reviews recent literature on TCM in AD treatment,summarizes the structure of AMPK and its mechanisms in AD,and presents the current status of research on TCM regulation of the AMPK signaling pathway for AD prevention and treatment,aiming to provide a theoretical foundation and new insights for AD management.
