摘要
目的探究金花茶叶提取物对小鼠酒精性肝损伤的保护作用。方法将60只雄性昆明小鼠随机分为对照组、模型组、低剂量组(0.75 g生药/kg)、中剂量组(1.50 g生药/kg)、高剂量组(3.00 g生药/kg),每组12只。对照组和模型组灌胃纯水,各提取物组灌胃不同剂量的提取物(20 mL/kg)。5 h后,除对照组外,其余各组均给予40%乙醇(10 mL/kg),连续处理14 d。末次给药后禁食16 h,再给予20 mL/kg乙醇。测定小鼠血清生化指标及肝匀浆抗氧化标志物(超氧化物歧化酶、谷胱甘肽、丙二醛、蛋白质羰基)及炎症细胞因子白介素-1β、白介素-6和肿瘤坏死因子-α的水平。采用苏木精-伊红(hematoxylin-eosin,HE)染色观察小鼠肝脏病理学变化。结果与模型组相比,高剂量组血清中甘油三酯下降21.76%、谷丙转氨酶下降15.88%、谷草转氨酶下降12.23%、肿瘤坏死因子-α水平下降37.17%,高剂量组肝脏中超氧化物歧化酶的含量升高14.96%、丙二醛含量下降17.41%,中、高剂量组谷胱甘肽的含量分别升高18.32%、24.10%,中、高剂量组蛋白质羰基含量分别下降15.38%、19.33%,中、高剂量组血清白介素-1β分别下降43.27%、49.72%,低、中、高剂量组白介素-6含量下降25.06%、28.30%、32.59%(P<0.05)。HE染色显示中、高剂量组中的肝细胞水肿、气球样变现象得到改善。结论金花茶叶提取物对酒精诱导的小鼠肝损伤有较好的保护作用,其作用可能与抗氧化和抗炎机制有关。本研究为金花茶的深入开发与利用提供实验依据。
Objective To investigate the protective effects of Camellia nitidissima leaves extract on alcoholic liver injury in mice.Methods The 60 male Kunming mice were randomly divided into control group,model group,收稿日期:2025-09-25基金项目:广西壮族自治区药监局药品安全科研项目(桂药监办函〔2022〕307号)第一作者:莫玉焕(1991—),女,硕士,工程师,主要研究方向为药检药理毒理研究。E-mail:1449173541@qq.com*通信作者:田婧(1988—),女,工程师,主要研究方向为药检药理毒理研究。E-mail:215941053@qq.com 76食品安全质量检测学报第17卷low-dose group(0.75 g crude drug/kg),medium-dose group(1.50 g crude drug/kg)and high-dose group(3.00 g crude drug/kg)(n=12 per group).The control and model groups were administered pure water by gavage,while extract groups received different doses of extract(20 mL/kg).After 5 hours,40%elthanol(10 mL/kg)was administered to all groups except the control group for 14 consecutive days.After fasting for 16 hours following the last administration,20 mL/kg ethanol was administered.Serum biochemical indicators,liver homogenate antioxidant markers(superoxide dismutase,glutathione,malondialdehyde,protein carbonyl)and inflammatory cytokines(interleukin-1β,interleukin-6 and tumor necrosis factor-α)were analyzed.The pathological changes of liver in mice were observed by hematoxylin-eosin(HE)staining.Results Compared with the model group,the serum triglyceride,alanine aminotransferase,aspartate aminotransferase and tumor necrosis factor-αlevels in the high dose group decreased by 21.76%,15.88%,12.23%and 37.17%,respectively.The content of superoxide dismutase in the liver of the high dose group increased by 14.96%,and the content of malondialdehyde decreased by 17.41%.The content of glutathione in the middle and high dose groups increased by 18.32%and 24.10%,respectively.The protein carbonyl content in the middle and high dose groups decreased by 15.38%and 19.33%,respectively.The serum interleukin-1βin the middle and high dose groups decreased by 43.27%and 49.72%,respectively.The interleukin-6 content in the low,middle and high dose groups decreased by 25.06%,28.30%and 32.59%,respectively(P<0.05).HE staining showed that hepatocyte edema and ballooning were improved in the medium and high dose groups.Conclusion Camellia nitidissima leaves extract has a good protective effect on alcoholic liver injury in mice,which may be related to antioxidant and anti-inflammatory mechanisms.This study provides experimental basis for the in-depth development and utilization of Camellia nitidissima.
