金花茶和银杏叶对2-乙基亚硝胺致大鼠肝癌前病变抑制作用的初步研究 被引量:14

摘要 目的探讨5%的金花茶叶和5%的金花茶浓缩液及5%银杏叶对DEN致肝癌抑制作用。方法应用广西医科大学附属肿瘤医院病理研究室建立的2-乙基亚硝胺(DEN)致大鼠作用的短期体内实验模型,用DEN作为肝癌的启动剂(200μg/kg静脉注射,只用1次),然后喂食含0.015%的2-AAF(2-乙酰氨基芴)饲料2周,加大部分肝切除,作为选择性促进程序,使被DEN启动的肝细胞迅速增生成癌前病灶。各实验组大鼠从实验一开始即喂食含5%的金花茶叶、5%金花茶浓缩液和5%银杏叶饲料,对照组则喂食基础饲料。大鼠于停饲2-AAF饲料3天后处死。肝脏按规定部位取材,做γ—谷氨酰转肽酶(γ-GT)染色,用我们自己设计的计算机软件计算肝癌前病变γ-GT阳性灶,算出平均每cm2肝面积中γ-GT阳性灶的数目、面积。结果实验组γ-GT阳性灶的数目、面积均明显少于和小于对照组。结论5%金花茶叶、5%的金花茶浓缩液和5%银杏叶均有抑制DEN致大鼠肝癌作用,且银杏叶具有抵抗DEN对大鼠的毒性作用。 Objective Using a short-term in vivo laboratory model of murine liver cancer induced with diethylnitrosamine (DEN) ,we inspected the impact of leaves of Camellia chrysantha and Ginkogo biloba in 5% concentration as well as 5% concentrated solution of C. chrysantha on induction of liver cancer induced with DEN. Methods Laboratory Wistar rats, all being male and SPF grade, were administered with the liver cancer initiator DEN (200ug/kg,ip, once only), then with murine food containing 0. 015% 2-AAF (2-Aeetaminoflueren) for 2 weeks,following an excision of the augmented parts as a selective promoting program that speeded up the proliferation of precancerous loci from the initiated hepatocytes. On the beginning of this experiment, animal food containing leaves of Camellia chrysantha and Ginkogo biloba in 5% concentration as well as 5% concentrated solution of C. chrysantha were given to the trail rats. Normal food was given to the control rats. All rats were killed 3 days after the end of feeding of 2-AAF-contained food. Targeting part of liver were collected for .γ-glutamyltranspeptidase (γ-GT) staining, and numbers and areas of γ-GT positive foci per em2 of the liver area were calculated using a software designed by us. Results Numbers and areas of γ-GT positive foei of the trail are significantly less than those of the control. Conclusion Leaves of Camellia chrysantha and Ginkogo biloba in 5% concentration as well as 5% concentrated solution of C. ehrysantha inhibit DEN-induction to liver cancer. Ginkogo biloba in 5% concentration also has activity against toxicity of DEN to liver.
