摘要
试验旨在探究饲料中添加槲皮素对鲫鱼幼鱼生长性能、肌肉营养成分、肝脏抗氧化酶活性和免疫相关基因表达量的影响。选取300尾初始体重为(4.82±2.10)g的健康鲫鱼幼鱼,随机分为4组,每组3个重复,每个重复25尾。对照组(CON组)饲喂基础饲料,LQE组、MQE组和HQE组分别在基础饲料中添加50、100和200 mg/kg槲皮素。预试期7 d,正式试验期56 d。结果显示,与CON组相比,HQE组增重率和特定生长率显著提高(P<0.05),肝体比显著降低(P<0.05)。LQE组、MQE组和HQE组超氧化物歧化酶(SOD)活性显著提高(P<0.05),丙二醛(MDA)含量显著降低(P<0.05);LQE组过氧化氢酶(CAT)和谷丙转氨酶(ALT)活性显著提高(P<0.05);MQE组CAT和碱性磷酸酶(AKP)活性显著提高(P<0.05);HQE组酸性磷酸酶(ACP)、AKP、ALT、谷草转氨酶(AST)和谷胱甘肽过氧化物酶(GSH-Px)活性显著降低(P<0.05)。HQE组肿瘤坏死因子-α(TNF-α)、白细胞介素-8(IL-8)、转化生长因子-β1(TGF-β1)、白细胞介素-10(IL-10)和CXC趋化因子受体1(CXCR1)基因相对表达量均显著提高(P<0.05)。组织学观察显示,MQE组鲫鱼肝脏无不良影响;HQE组鲫鱼肝脏细胞受到轻微损伤。研究表明,槲皮素添加量为200 mg/kg时,对鲫鱼生长和发育的促进效果较好;添加量为100 mg/kg时,能够增强肝脏抗氧化能力,对肝脏保护效果较好。
This experiment aimed to investigate the effects of dietary quercetin supplementation on the growth performance,muscle nutritional composition,hepatic antioxidant enzyme activities,and immune-related gene expression levels in juvenile Carassius auratus.A total of 300 healthy juvenile Carassius auratus with an initial body weight of(4.82±2.10)g were randomly divided into four groups,with three replicates per group and 25 fish per replicate.The control group(group CON)was fed a basal diet,while groups LQE,MQE,and HQE were fed the basal diet supplemented with 50,100,and 200 mg/kg quercetin,respectively.The pre-test period was seven days,and the formal test period was 56 days.The results showed that compared with group CON,the weight gain rate and specific growth rate in group HQE were significantly increased(P<0.05),and the hepatosomatic index was significantly decreased(P<0.05).Superoxide dismutase(SOD)activity in groups LQE,MQE,and HQE was significantly increased(P<0.05),and malondialdehyde(MDA)content was significantly decreased(P<0.05).Catalase(CAT)and alanine aminotransferase(ALT)activities in group LQE were significantly increased(P<0.05).CAT and alkaline phosphatase(AKP)activities in group MQE were significantly increased(P<0.05).Acid phosphatase(ACP),AKP,ALT,aspartate aminotransferase(AST),and glutathione peroxidase(GSH-Px)activities in group HQE were significantly decreased(P<0.05).The relative expression levels of tumor necrosis factor-α(TNF-α),interleukin-8(IL-8),transforming growth factor-β1(TGF-β1),interleukin-10(IL-10),and CXC chemokine receptor 1(CXCR1)in group HQE were significantly increased(P<0.05).Histological observation indicated that group MQE had no adverse effects on the liver of Carassius auratus,whereas group HQE caused slight damage to hepatocytes.The study shows that a quercetin supplementation level of 200 mg/kg yields the optimal promoting effect on the growth and development of Carassius auratus,while a level of 100 mg/kg enhances hepatic antioxidant capacity and provides the optimal protective effect on the liver.
