摘要
为研究低温胁迫对水稻(Oryza sativa L.)幼苗根系活性氧代谢及其相关基因表达的影响,本研究以苗期耐冷性不同的2个水稻品种‘湘早籼45号’和‘三七十萝’为材料,通过人工模拟低温胁迫,检测和分析低温胁迫对水稻根系形态、根系活力、根系酶活性、根系分泌物含量、根系渗透调节物质及其相关基因表达之间的关系。结果表明,与适温对照相比,低温胁迫下幼苗总根长、根表面积、根直径、根总体积和根系活力下降,且随着处理时间的延长降低越明显,对照品种‘湘早籼45号’幼苗的降幅大于耐冷品种‘三七十萝’;低温胁迫下根系过氧化氢酶(catalase,CAT)活性、过氧化物酶(peroxidase,POD)活性表现为先增加后降低的趋势,根系相对电导率、MDA含量、超氧阴离子含量与H_(2)O_(2)含量增加,且耐冷品种‘三七十萝’根系CAT活性、POD活性、超氧阴离子含量、H_(2)O_(2)含量、相对电导率、MDA含量变幅小于对照品种‘湘早籼45号’;低温胁迫下根系渗透调节物质脯氨酸(Pro)含量增加,可溶性蛋白含量先增加后降低,且‘湘早籼45号’的变幅大于‘三七十萝’;低温胁迫下根系活性氧代谢相关基因(POD,CAT,APX,MDAR)的表达水平呈现降低趋势,耐冷品种‘三七十萝’根系的活性氧代谢相关基因表达水平高于‘湘早籼45号’。本研究对开展水稻耐冷性研究,对促进水稻生产持续、安全、稳步发展具有深远意义。
In order to study the impacts of low temperature stress on reactive oxygen species metabolism and related gene expression in roots of rice(Oryza sativa L.)seedlings,two rice varieties'Xiangzaoxian 45'and'Sanqishiluo'with different cold tolerance at seedling stage were used as materials.The relationship between root morphology,root activity,root enzyme activity,root exudate content,root osmotic regulatory substances and related gene expression under low temperature stress was detected and analyzed.The results indicated that,compared with the optimum temperature control,the total root length,root surface area,root diameter,root volume,and root activity of seedlings decreased under low temperature stress,and the extent of reduction became more pronounced with prolonged treatment time.The decrease rate of seedlings of control variety'Xiangzaoxian 45'was greater than that of cold-tolerant variety'Sanqishiluo'.Under low temperature stress,the activities of CAT and POD in roots increased first and then decreased,while the relative conductivity,MDA content,superoxide anion content and H_(2)O_(2) content in roots increased.The variation of CAT activity,POD activity,superoxide anion content,H_(2)O_(2) content,relative conductivity and MDA content in roots of the cold-tolerant cultivar'Xiangzaoxian 45'was smaller than that of the control cultivar'Xiangzaoxian 45'.Under low temperature stress,proline content of root osmotic regulation substance increased,soluble protein content increased first and then decreased,and the variation range of'Xiangzaoxian 45'was larger than that of'Sanqishiluo'.The expression levels of POD,CAT,APX and MDAR generally showed a decreasing trend.The expression levels of POD,CAT,APX and MDAR genes in roots of the cold-tolerant cultivar'Sanqishiluo'were higher than those of'Xiangzaoxian 45'.This study is of far-reaching significance to the research on cold tolerance of rice and to promote the sustainable,safe and steady development of rice production.
