电针对肥胖大鼠骨骼肌组织ADRB2、NR4A3表达的影响

Effects of electroacupuncture on ADRB2 and NR4A3 expressions in skeletal muscle of obese rats

摘要 目的观察肥胖对骨骼肌葡萄糖摄取的影响以及电针对高脂饮食诱导的肥胖大鼠骨骼肌组织中β2肾上腺素受体(ADRB2)、核受体亚家族4成员3(NR4A3)、葡萄糖转运蛋白4(GLUT4)表达的影响。方法从30只Wistar雄性大鼠中随机挑选10只作为正常组,给予标准饲料喂养;剩余的20只大鼠给予高脂饲料喂养建立肥胖模型。将造模成功的大鼠按照随机数字表法分为模型组和电针组,每组10只。使用韩氏电针仪,对电针组大鼠中脘-关元、足三里-丰隆(组成双回路刺激系统)施加2 Hz连续波,电流强度1 mA,每次干预20 min,每周3次,持续干预8周。测量大鼠干预前后体重、空腹血糖(FPG)、餐后2 h血糖(2hPG),全自动生化分析仪检测干预8周后糖化血红蛋白(HbA1c)及血清总胆固醇(TC)、三酰甘油(TG)、游离脂肪酸(FFA)水平,Western blot法检测骨骼肌组织中ADRB2和GLUT4蛋白表达情况,荧光定量PCR法检测骨骼肌组织中ADRB2、NR4A3、GLUT4 mRNA表达情况,免疫组化法检测骨骼肌组织中ADRB2阳性表达情况。结果干预前和干预8周后,模型组和电针组大鼠体重、FPG、2hPG均明显高于同期正常组(P均<0.05);干预8周后,电针组大鼠体重、FPG、2hPG均明显低于模型组(P均<0.05)。与正常组比较,模型组大鼠血清HbA1c、TG、FFA水平均明显升高(P均<0.05),骨骼肌组织中ADRB2和GLUT4蛋白相对表达量,ADRB2、NR4A3、GLUT4 mRNA相对表达量和ADRB2阳性面积占比均明显降低(P均<0.05);与模型组比较,电针组大鼠血清HbA1c、TG、FFA水平均明显降低(P均<0.05),骨骼肌组织中ADRB2和GLUT4蛋白相对表达量,ADRB2、NR4A3、GLUT4 mRNA相对表达量和ADRB2阳性面积占比均明显升高(P均<0.05)。结论电针能促进高脂饮食喂养肥胖大鼠骨骼肌葡萄糖摄取,其作用机制可能与上调骨骼肌中ADRB2、NR4A3的表达,从而促进GLUT4表达有关。 Objective It is to observe the effect of obesity on skeletal muscle glucose uptake and the effects of electroacupuncture(EA)on the expressions ofβ2 adrenergic receptor(ADRB2),nuclear receptor subfamily 4 group A member 3(NR4A3)and glucose transporter type 4(GLUT4)in skeletal muscle of high-fat diet(HFD)-induced obese rats.Methods Thirty male Wistar rats were taken,in which 10 rats were randomly selected as normal group and fed with standard diet;the 20 remaining rats were fed with high-fat diet to establish models of obesity.The successfully modeled rats were divided into normal model group and EA group,with 10 rats in each group.The rats in the EA group were treated with Han’s electroacupuncture device at Zhongwan(CV12)-Guanyuan(CV4),Zusanli(ST36)-Fenglong(ST40)(forming a dual-circuit stimulation system),2 Hz continuous wave with an current intensity of 1 mA,20 min per time,3 times weekly,continuously treated for 8 weeks.The body weight,fasting plasma glucose(FPG),2-hour postprandial glucose(2hPG)of the rats were measured before and after treatment,the levels of glycated hemoglobin(HbA1c),serum total cholesterol(TC),triglycerides(TG)and free fatty acids(FFA)were measured by fully automated biochemical analyzer after 8 weeks of treatment,the protein expressions of ADRB2 and GLUT4 in skeletal muscle tissue were detected by Western blot,the mRNA expressions of ADRB2,NR4A3 and GLUT4 in skeletal muscle tissue were detected by fluorescent quantitative PCR,the positive expression of ADRB2 in skeletal muscle tissue was detected by immunohistochemistry.Results Before intervention and after 8 weeks of intervention,the body weight,FPG,and 2hPG of rats in the model group and EA group were significantly higher than those in the normal group at the same time point(all P<0.05);after 8 weeks of intervention,the body weight,FPG,and 2hPG of rats in the EA group were significantly lower than those in the model group(all P<0.05).Compared with the normal group,the serum levels of HbA1c,TG,and FFA were significantly increased,while the relative protein expressions of ADRB2 and GLUT4,relative mRNA expressions of ADRB2,NR4A3 and GLUT4 and the ADRB2-positive area fraction in skeletal muscle tissue of rats were all significantly reduced in the model group(all P<0.05).Compared with the model group,the serum levels of HbA1c,TG,and FFA were significantly decreased,while the relative protein expressions of ADRB2 and GLUT4,relative mRNA expressions of ADRB2,NR4A3 and GLUT4 and the ADRB2-positive area fraction in skeletal muscle tissue of rats were all significantly increased in the EA group(all P<0.05).Conclusion Electroacupuncture can promote glucose uptake in skeletal muscle of high-fat diet-fed obese rats,the mechanism may be related to upregulating expressions of ADRB2 and NR4A3 in skeletal muscle,thereby promoting expression of GLUT4.
