摘要
目的:筛选优化复方余甘子-三白纳米乳(CPA-NE)的制备工艺,并对其抗氧化活性进行评价。方法:应用伪三元相图法结合单因素试验,以平均粒径为指标通过Box-Behnken响应面设计优化CPA-NE的制备工艺,并对其外观形貌、粒径分布、包封率、稳定性和抗氧化活性等进行评价。结果:优化的处方条件为油相4%,吐温8018%,无水乙醇6%,水相72%;CPA-NE包封率为(90.16±2.6)%,粒径为(14.33±0.53)nm,PDI为(0.06±0.01),Zeta电位为(-10.29±1.10)m V,p H值为(6.53±0.01),黏度为(41.51±0.30)m Pa·s^(−1),浊度为(5.17±0.19)cm^(−1)。CPA-NE外观澄清透明,为O/W型纳米乳,形态呈规整的类球形,稳定性好。体外经皮释放研究结果:CPA-NE累计释放量高达843.37μg·cm^(−2),具有良好的药物经皮促渗作用。抗氧化实验中CPA-NE清除DPPH与ABTS自由基性能显著。结论:经伪三元相图法结合响应面法优化得到的CPA-NE稳定性好,具有良好的经皮促透作用和抗氧化活性,可为复方中药纳米乳液护肤品的研制及应用提供参考。
Objective:To screen and optimise the preparation process of compound phyllanthus emblica Sanbai Chinese medicine nanoemulsion(CPA NE)and evaluate its antioxidant activity.Methods:The preparation process of CPA NE was optimised by Box Behnken response surface design by applying the pseudo ternary phase diagram experiment combined with a single factor experimentation using the average particle size as an indicator,and evaluate its appearance,particle size distribution,encapsulation rate,stability and antioxidant activity.Results:The optimized prescription conditions were 4%oil phase,18%Tween 80,6%anhydrous ethanol and 72%aqueous phase;the CPA NE encapsulation rate was(90.16±2.6)%,the particle size was(14.33±0.53)nm,the PDI was 0.06±0.01,the zeta potential was(-10.29±1.10)mV,the pH was 6.53±0.01,the viscosity was(41.51±0.30)mPa·s^(−1),and turbidity was(5.17±0.19)cm^(−1).CPA NE is a clear and transparent O/W nano-emulsion with regular spherical shape and good stability.The in vitro transdermal release study showed that the cumulative release of CPA-NE was as high as 843.37μg`cm^(−2),which had a good transdermal permeation promotion effect.The scavenging performance of CPA NE in the antioxidant assay was significant in scavenging DPPH and ABTS radicals.Conclusion:The CPA NE optimized by pseudo-ternary phase diagram method combined with response surface method is stable and has good transdermal permeation promotion and antioxidant activity,which can provide a reference for the development and application of compounded traditional Chinese medicine nanoemulsion skin care products.
