紫铆因对牛冠状病毒的体外抗病毒作用

Antiviral activity of butein on bovine coronavirus in vitro

摘要 牛冠状病毒(BCoV)是引起犊牛腹泻和牛呼吸道疾病的主要病原之一,世界性流行,对畜牧业造成严重经济损失。研究利用RT-qPCR、蛋白免疫印记法(Western blot)、IFA和TCID50等技术检测紫铆因体外对BCoV复制的影响。结果表明:紫铆因可有效抑制BCoV复制,降低细胞内BCoV N基因表达量、BCoV N蛋白表达量和病毒滴度。半数有效浓度(EC_(50))和选择指数(SI)分别是5.25 mg·L^(-1)和4.63;利用RT-qPCR检测显示,紫铆因抑制BCoV复制阶段,对BCoV吸附、内化和出芽过程无影响。研究结果认为,紫铆因在细胞水平上具有抗病毒作用,可为开发抗BCoV药物提供参考。 Bovine coronavirus(BCoV)is one of the main pathogens causing diarrhea in calves and respiratory diseases in cattle,it has a global prevalence and causes significant economic losses to the livestock industry.Techniques,such as RT-qPCR,Western blot,immuno-fluorescence assay and TCID50 were employed to investigate the effect of butein on BCoV replication in vitro.The results showed that butein effectively suppressed BCoV replication by reducing the intracellular expression levels of the BCoV N gene,BCoV N protein and viral titers.The half-maximal effective concentration(EC_(50))and selectivity index(SI)were 5.25 mg·L^(-1) and 4.63,respectively.Further analysis using RT-qPCR to assess the impact of butein on different stages of viral infection revealed that butein specifically inhibited the replication stage of BCoV,but had no significant effect on the adsorption,internalization,or budding processes.This study clarified that butein had antiviral effects at the cellular level,laying the foundation for the development of anti-BCoV drugs.
