摘要
为提高药用植物对不同胁迫的适应性,以黄花铁线莲种子和幼苗为研究对象,探究盐胁迫和干旱胁迫对种子萌发和幼苗生长及生理特性的影响。结果表明,盐胁迫时,随着NaCl浓度的升高,种子的发芽率、发芽势、发芽指数均呈下降趋势,而活力指数和萌发后的形态指标在低浓度(25 mmol/L)时有显著促进作用。随着NaCl浓度的变化,幼苗在25 mmol/L时叶片长势较对照最佳,鲜重则呈逐渐下降的变化趋势,幼苗CAT活性呈升-降-升的趋势,在100 mmol/L时最高;POD活性和MDA含量逐渐下降,SOD活性呈升-降-升的趋势,在25 mmol/L时最高,脯氨酸含量呈先升高后降低的趋势,在75 mmol/L时最高。干旱胁迫时,随着PEG浓度的升高,种子的发芽率、发芽势均在8%时最高,而发芽指数、活力指数分别在2%和4%时最高,萌发后的形态指标在4%时最佳。幼苗的生长指标均随着PEG浓度的增加逐渐下降,CAT活性呈升-降-升的趋势,在2%时最高,MDA与脯氨酸含量呈先升高后降低的趋势,均在8%时最高。研究表明,黄花铁线莲对低浓度的盐和干旱胁迫具有一定的耐受能力。
To enhance the adaptability of medicinal plants to different stresses,taking the seeds and seedlings of Clematis intricata as the research objects,the effects of salt stress and drought stress on seed germination,seedling growth and physiological characteristics were explored.The results showed that under salt stress,with the increase of NaCl concentration,the germination rate,germination potential and germination index of seeds all showed a downward trend,while the vitality index and morphological indicators after germination had a significant promoting effect at a low concentration(25 mmol/L).With the change of NaCl concentration,the leaf growth of the seedlings was the best at 25 mmol/L compared with the control,while the fresh weight showed a gradually decreasing trend.The CAT activity of the seedlings showed an upward-decreasing-upward trend,and was the highest at 100 mmol/L.The activity of POD and the content of MDA gradually decreased.The activity of SOD showed an upward-downward-upward trend,reaching the highest at 25 mmol/L.The content of proline showed a trend of first increasing and then decreasing,reaching the highest at 75 mmol/L.Under drought stress,with the increase of PEG concentration,the germination rate and germination potential of seeds were the highest at 8%,while the germination index and vitality index were the highest at 2%and 4%respectively.The morphological index after germination was the best at 4%.The growth indicators of the seedlings gradually decreased with the increase of PEG concentration.The CAT activity showed a trend of rising-decreasing-rising,reaching the highest at 2%.The contents of MDA and proline showed a trend of first increasing and then decreasing,both reaching the highest at 8%.Studies showed that the Clematis inteicata had a certain tolerance to low salt concentrations and drought stress.
