摘要
本研究基于Fe_(3)O_(4)@Au复合纳米材料光热效应与类过氧化物酶活性,建立了一种双模式信号放大的适配体试纸条方法,用于鼠伤寒沙门菌的快速检测。在最优条件下,通过类过氧化物酶催化显色,该方法的目视检出限为10^(4)CFU/mL;利用Fe_(3)O_(4)@Au的光热效应,通过建立检测线上温差(ΔT)与细菌浓度之间的定量标准曲线,可在10^(4)~10^(8)CFU/mL范围内实现线性检测,最低检测限达3.02×10^(2)CFU/mL。该方法对鼠伤寒沙门菌具有高特异性,与乙型副伤寒沙门菌、单核细胞增生李斯特菌、大肠埃希菌O157:H7、金黄色葡萄球菌均无交叉反应。试纸条具备良好的贮藏稳定性,并能够成功应用于牛奶、鸡胸肉和猪肉等实际样本中鼠伤寒沙门菌的检测,回收率在80.54%~104.35%之间。本研究建立的方法集快速、便捷、准确等优势于一体,成功实现了复杂食品基质中鼠伤寒沙门菌的快速检测,有望为食品安全现场快速筛查提供有效的新策略。
In this study,a novel dual-mode signal amplification aptamer-based test strip method that utilizes the photothermal effect and peroxidase-like activity of Fe_(3)O_(4)@Au composite nanomaterials was developed for the rapid detection of Salmonella typhimurium.Under optimal conditions,a visual detection limit of 10^(4)CFU/mL was achieved based on peroxidase-like catalytic coloration.By utilizing the photothermal effect of Fe_(3)O_(4)@Au,a quantitative standard curve was established by plotting the temperature change(ΔT)against bacterial concentration.A linear detection range from 10^(4)to 10^(8)CFU/mL was achieved,with a lowest detection limit of 3.02×10^(2)CFU/mL.The method demonstrated high specificity for S.typhimurium,and showed no cross-reactivity with Salmonella Paratyphi B,Listeria monocytogenes,Escherichia coli O157:H7,or Staphylococcus aureus.Furthermore,the test strip exhibited satisfactory storage stability.It was successfully applied to detect S.typhimurium in real samples,including milk,chicken breast,and pork.The recovery rates ranged from 80.54%to 10^(4).35%.With its advantages including rapidity,simplicity,and accuracy,this method can effectively detect S.typhimurium in complex food matrices.It represents a promising strategy for rapid on-site food safety screening.
