摘要
苯甲酸作为常用防腐剂,其含量检测对保障食品安全具有重要意义。本研究针对国家标准GB5009.28-2016中苯甲酸的色谱分析结果存在基质干扰影响检测准确性的不足,对前处理方法和液相色谱流动相等进行了方法优化研究,采用亚铁氰化钾(150 g/L)-乙酸锌(300 g/L)沉淀净化样品,用甲醇-水体系(比例为60∶40(v/v))提取目标物,以甲醇-0.005 mol/L乙酸铵缓冲液(pH值4.5±0.1)为流动相,实现了苯甲酸的准确定量。方法检出限为0.015 mg/kg,线性范围为0.05~10.0 mg/kg,加标回收率在93.4%~98.7%之间。该方法具有前处理简单、重现性好、基质干扰小等特点,满足食品中苯甲酸的日常检测要求,可为食品质量安全监测提供可靠的分析方法。
Benzoic acid,as a commonly used preservative,its content detection is of great significance for ensuring food safety.This study aimed at the deficiency of matrix interference in the chromatographic analysis results of benzoic acid in the national standard GB 5009.28-2016,which affected the detection accuracy.A method optimization study was conducted on the pretreatment method and liquid chromatography flow phase.Potassium ferrocyanide(150g/L)-zinc acetate(300g/L)precipitation was used to purify the samples.The target substance was extracted using a methanol-water system(with a ratio of 60∶40(v/v)),and the mobile phase was methanol-0.005 mol/L ammonium acetate buffer(pH value 4.5±0.1),achieving accurate quantification of benzoic acid.The detection limit of the method was 0.015mg/kg,the linear range was 0.05~10.0mg/kg,and the spiked recovery rate was between 93.4%and 98.7%.This method features simple pretreatment,good reproducibility and minimal matrix interference.It meets the daily detection requirements of benzoic acid in food and can provide a reliable analytical method for food quality and safety monitoring.
