基于广泛靶向代谢组学及网络药理学研究大黄的次生代谢物与药理作用差异

Differences in secondary metabolites and pharmacological effects of RHEI RADIX ET RHIZOMA revealed by widely targeted metabolomics and network pharmacology

摘要 目的:利用广泛靶向代谢组学及网络药理学,系统探索大黄鲜切饮片X1、晾干饮片X2及成品饮片C3间的次生代谢物差异,并深入剖析这些差异代谢物与其药理作用间的关系。方法:采用UPLC-MS/MS技术对大黄样品进行次生代谢物的高效筛选与鉴定,筛选差异成分;采用网络药理学分析差异成分对药理作用的具体影响,揭示潜在的作用靶点及生物学过程。结果:共检测出1 046种化学成分,涵盖核苷酸类、醌类、鞣质类等多类别。不同样品的次生代谢物组成上存在显著差异,表现为C3与X1存在489种成分下调、263种成分上调;C3与X2有445种成分下调、222种成分上调;X2与X1有367种成分下调、257种成分上调。网络药理学分析揭示X1与X2虽共享部分核心靶点,但也存在独特的靶点差异(如PIK3R1、PTK2、ITGB1等),这些差异靶点与细胞钙离子平衡、离子平衡、MAP激酶活性调节等生物学过程密切相关,是导致两者药理作用差异的关键因素。结论:该实验揭示了大黄不同处理状态下次生代谢物的显著差异及其与药理作用之间的复杂关系。 OBJECTIVE To systematically compare the secondary metabolites of fresh-cut(X1),air-dried(X2),and pro⁃cessed(C3)RHEI RADIX ET RHIZOMA slices using widely targeted metabolomics,and to relate these differences to pharma⁃cological effects using network pharmacology.METHODS UPLC-MS/MS was employed to comprehensively profile and anno⁃tate secondary metabolites across the three processing states.Differential metabolites between groups were identified,and net⁃work pharmacology was applied to infer putative targets and associated biological processes.RESULTS In total,1046 metabo⁃lites were detected,spanning classes such as nucleotides,quinones,and tannins.Substantial compositional differences were observed:relative to X1,C3 had 489 downregulated and 263 upregulated metabolites;relative to X2,C3 had 445 downregulated and 222 upregulated metabolites;and X2 versus X1 showed 367 downregulated and 257 upregulated metabolites.Network phar⁃macology indicated that X1 and X2 share several core targets but also differ in key targets(e.g.,PIK3R1,PTK2,ITGB1)asso⁃ciated with ion homeostasis(including calcium homeostasis)and regulation of MAP kinase activity.These target differences likely contribute to the pharmacological divergences among processing states.CONCLUSION Processing state markedly alters the sec⁃ondary metabolite profiles of Rhei Radix et Rhizoma and is linked to distinct,target-related pharmacological effects.
