舞草属叶绿体基因组特征及系统发育分析

摘要 舞草属(Codariocalyx Hassk)有舞草[C.motorius(Houttuyn)H.Ohashi]和圆叶舞草[C.gyroides(Roxburgh ex Link)Hasskarl]2种,其中舞草的侧生小叶可以进行上下摆动或旋转运动。为探究舞草属的叶绿体基因组特征及系统发育关系,本研究通过Illumina平台对其进行高通量测序,并运用GetOrganelle、CPStools等对舞草属叶绿体基因组基本特征、重复序列、序列差异、密码子偏好性及系统发育关系进行分析。结果表明:(1)舞草和圆叶舞草的叶绿体基因组总长度分别为149381 bp和149358 bp,总GC值均为35%,基因128个,其中,蛋白编码基因83个,密码子偏好以A/U结尾;(2)SSR序列在舞草中以T和AT为主,在圆叶舞草中则主要为A和AT,其中仅有圆叶舞草具有五碱基重复(TTATA);而散在重复序列则均以回文重复和正向重复为主;(3)序列差异分析筛选得到4个高变异区域(trnP-UGG_psaJ、psbD_trnTGGU、trnY-GUA_trnD-GUC和psaI_ycf4);(4)舞草和圆叶舞草聚为单系支,与山蚂蝗亚族内Desmodium group物种聚为一支,并与假地豆属的系统发育关系最近。 Codariocalyx Hassk.contains two species,C.motorius and C.gyroides.The lateral leaflets of C.motorius can swing up and down nearly 180°or rotate 360°.To investigate the chloroplast genome characteristics and phylogenetic relationships of the genus Codariocalyx,this study was based on high-throughput sequencing using the Illumina platform.Bioinformatics tools such as GetOrganelle and CPStools were employed to assemble and analyze the chloroplast genomes.This study focused on their structural features,repeat sequences,sequence divergence,codon usage bias,and phylogenetic placement within subtribe Desmodiinae.The results showed:(1)The total lengths of the chloroplast genomes of C.motorius and C.gyroides were 149381 bp and 149358 bp,respectively,with a total GC content of 35%.A total of 128 genes were identified,including 83 protein-coding genes.(2)SSR sequences in C.motorius were predominantly composed of T and AT repeats,whereas in C.gyroides,A and AT repeats dominated.Notably,only C.gyroides contained a pentanucleotide repeat(TTATA).Dispersed repetitive sequences in both species mainly consisted of palindromic repeats and direct repeats.(3)Sequence divergence analysis identified four highly variable regions(trnP-UGG_psaJ,psbD_trnT-GGU,trnY-GUA_trnD-GUC and psaI_ycf4).(4)C.motorius and C.gyroides formed a single branch,belonging to the Desmodium group of the tribe Desmodieae,and showed the closest phylogenetic relationship with Grona.This study provides a theoretical basis for species identification,genetic diversity research,and resource development and utilization of C.motorius and C.gyroides.
