CDH11基因过表达载体构建及其对牛成肌细胞增殖和分化的作用

Construction of CDH11 Gene Overexpression Vector and Its Effect on Proliferation and Differentiation of Bovine Myoblasts

摘要 通过在牛成肌细胞中过表达钙粘蛋白11(Cadherin-11,CDH11)基因并利用EDU、CCK-8、免疫荧光和实时荧光定量PCR(qRT-PCR)等技术,探讨CDH11基因对牛成肌细胞增殖与分化的作用。结果表明,过表达CDH11基因显著提高牛成肌细胞的EDU阳性率及450 nm波长下的吸光度(P<0.05),显著增强周期蛋白依懒性激酶2(Cyclin-Dependent Kinases,CDK2)和增殖细胞核抗原(Proliferating Cell Nuclear Antigen,PCNA)的表达水平(P<0.05)。在成肌细胞分化阶段,过表达CDH11基因显著减少了肌管的面积,显著抑制成肌分化相关基因成肌分化因子(Myogenic Diffeerentiation,MYOD)、肌细胞生成素(Myogenin,MYOG)和生肌调节因子5(Myogenic Regulatory Factors5,MYF5)的表达(P<0.05)。研究表明过表达CDH11基因能够促进牛成肌细胞的增殖,但对其分化具有抑制作用。 The role of cadherin-11(CDH11)gene in the proliferation and differentiation of bovine adult myoblasts was investigated by overexpressing CDH11 and using EdU,CCK-8,immunofluorescence and quantitative real-time PCR(qRT-PCR).The results showed that CDH11 overexpression significantly increased the EdU positivity and absorbance at 450 nm in bovine myogenic cells(P<0.05).The expression levels of cyclin-dependent kinase(CDK2)and Proliferating Cell Nuclear Antigen(PCNA)were significantly upregulated(P<0.05).During the myoblast differentiation stage,CDH11 overexpression significantly reduced the area of myotubes and markedly inhibited the myogenic differentiation-related genes myogenic differentiation(MYOD),myogenin(MYOG)and myogenic regulatory factors5(MYF5)expression(P<0.05).These findings indicate that CDH11 overexpression promotes the proliferation of bovine adult myoblasts but inhibits the differentiation of bovine adult myoblasts.
