基于等离激元共振效应的microRNA-451暗场原位检测

摘要 实现结直肠癌的无创早期诊断是一直亟待解决的问题.以miR-451作为结直肠癌检测的相关标志物,以单链DNA作为检测探针,以暗场显微镜为检测手段,以金纳米颗粒的LSPR效应为传感原理,以PDMS芯片为检测平台,将不同浓度的miR-451诱导修饰探针的金纳米颗粒的聚集量,转化为金纳米颗粒LSPR效应的散射峰偏移量,实现了对结直肠癌肿瘤潜在标志物miR-451的无创原位定量检测. To achieve noninvasive early diagnosis of colorectal cancer is an urgent problem to be solved.miR-451 was used as a marker for colorectal cancer detection,single-stranded DNA as a detection probe,dark field microscopy as a detection method,LSPR effect of gold nanoparticles as the sensing principle,PDMS chip as a detection platform.The accumulation of gold nanoparticles induced by different concentrations of miR-451 modified probes was converted into the scattering peak offset of the LSPR effect of gold nanoparticles,so as to achieve noninvasive in situ quantitative detection of miR-451,a poten-tial tumor marker of colorectal cancer.
