摘要
目的:探讨不育1号方(BY1F)通过调节肠道-睾丸轴治疗弱精子症(AZS)的机制。方法:采用雷公藤多苷(GTW)建立AZS小鼠模型。造模成功后随机分为正常组、模型组、BY1F低剂量组、BY1F中剂量组、BY1F高剂量组和左卡尼汀组,每组10只。干预4周后检测各组小鼠精子浓度与活力,通过苏木精-伊红(HE)染色、过碘酸-雪夫(PAS)染色和免疫组织化学(IHC)分析睾丸及结肠组织病理变化和闭锁蛋白(Occludin)与闭合小环蛋白-1(ZO-1)表达,利用16S rRNA测序分析肠道菌群(GM)结构及多样性。结果:与正常组比较,模型组小鼠精液质量显著下降(P<0.01),并伴有睾丸生精小管结构紊乱、生精细胞脱落、结肠黏膜损伤、杯状细胞数量减少(P<0.01),GM结构失调以及结肠与睾丸组织中Occludin和ZO-1表达下调(P<0.01)。BY1F干预能显著提升精子浓度与活力(P<0.05,P<0.01),改善病理损伤,增加杯状细胞数量(P<0.01),重塑GM结构,上调Occludin和ZO-1表达(P<0.01)。结论:BY1F通过调节GM结构,恢复肠道屏障和血睾屏障,改善GTW诱导的AZS模型小鼠的精液质量和睾丸组织病理学损伤,为中医药治疗男性不育提供了理论依据。
Objective:To investigate the mechanism by which Buyu No.1 Formula(BY1F)treats asthenozoospermia(AZS)through modulation of the gut-testis axis.Methods:An AZS mouse model was established using polyglycosides of Tripterygium wilfordii(GTW).After successful modeling,the mice were randomly divided into the normal group,model group,BY1F low-dose group,BY1F medium-dose group,BY1F high-dose group,and L-carnitine group,with 10 mice in each group.After 4 weeks of intervention,sperm concentration and motility were measured in each group.Hematoxylin-eosin(HE)staining,periodic acid-Schiff(PAS)staining,and immunohistochemistry(IHC)were used to analyze histopathological changes in the testicular and colonic tissues,as well as the expression of Occludin and zonula occludens-1(ZO-1).The structure and diversity of the gut microbiota(GM)were analyzed using 16S rRNA sequencing.Results:Compared with the normal group,the model group showed a significant decline in semen quality(P<0.01),along with disordered seminiferous tubule structure,shedding of spermatogenic cells,colonic mucosal injury,and a reduced number of goblet cells(P<0.01),as well as dysregulation of GM structure and downregulated expression of Occludin and ZO-1 in the colonic and testicular tissues(P<0.01).BY1F intervention significantly increased sperm concentration and motility(P<0.05,P<0.01),improved pathological damage,increased goblet cell number(P<0.01),reshaped GM structure,and upregulated Occludin and ZO-1 expression(P<0.01).Conclusion:BY1F improves semen quality and testicular histopathological injury in GTW-induced AZS model mice by regulating GM structure and restoring the intestinal barrier and blood-testis barrier,providing a theoretical basis for the application of traditional Chinese medicine in the treatment of male infertility.
