摘要
旨在为芝麻油市场监管提供技术支撑,建立了一种基于高效液相色谱技术同时测定芝麻油及芝麻调和油中8种特征香味物质含量的方法。样品(5 g)用10 mL甲醇超声提取20 min,经Phenomenex Gemini C6-Phenyl 110Å色谱柱(250 mm×4.6 mm,5μm)分离,柱温35℃,以乙腈-0.1%磷酸水溶液(体积比12∶88)为流动相进行等度洗脱,采用配备二极管阵列检测器的高效液相色谱仪进行检测,外标法定量。方法学考察结果表明,8种特征香味物质在0.5~10.0μg/mL质量浓度范围内线性关系良好,决定系数超过0.999,方法的检出限和定量限分别为0.4 mg/kg和1.0 mg/kg,加标回收率为84.00%~109.00%,精密度相对标准偏差为0.19%~1.68%。综上,该方法具有较高的灵敏度和精密度,能够准确、快速地同时检测芝麻油及芝麻调和油中8种特征香味物质。
In order to provide technical support for the market supervision of sesame oil,a method based on high performance liquid chromatography was established for the simultaneous determination of 8 characteristic aroma compounds in sesame oil and sesame blended oil.The samples(5 g)were extracted with 10 mL of methanol and ultrasonically extracted for 20 min.The extract were separated on a Phenomenex Gemini C6-Phenyl 110 A column(250 mm×4.6 mm,5μm),column temperature at 35℃.The mobile phase was acetonitrile-0.1%phosphoric acid aqueous solution(volume ratio 12∶88)for isocratic elution.Detection was carried out using a high performance liquid chromatography with diode array detector and external standard quantification.The results of the methodological investigation showed that the 8 characteristic aroma compounds had a good linear relationship within the mass concentration range of 0.5-10.0μg/mL,with determination coefficient exceeding 0.999.The detection limit and quantification limit of the method were 0.4 mg/kg and 1.0 mg/kg,respectively.The spiked recovery was 84.00%-109.00%,and the relative standard deviation for precision was 0.19%-1.68%.In summary,this method has high sensitivity and precision,and can accurately and quickly detect 8 characteristic aroma compounds in sesame oil and sesame blended oil simultaneously.
