温郁金对脑卒中后抑郁大鼠影响的实验研究

摘要 目的探讨温郁金抗脑卒中后抑郁(PSD)的作用及机制,为阐释中医“祛瘀生新”内涵提供研究基础。方法采用线栓大鼠大脑中动脉(MCAO)结合21天慢性不可预见性温和刺激(CUMS)方法制备PSD大鼠模型,并将大鼠分为假手术组、脑卒中组、PSD组、温郁金低剂量组(1.4 g生药/kg)、温郁金高剂量组(2.8 g生药/kg)和阳性药物组(氟西汀,20 mg/kg)。在应激前及应激后三个时间点采用Zea Longa评分来评价大鼠的神经功能缺损情况。采用HE染色观察大鼠海马区细胞形态结构变化;运用BrdU和Nestin免疫组织化学法染色评估海马区细胞增殖及神经干细胞再生情况。结果与假手术组对比,应激前和应激第7天,脑卒中组、PSD组、温郁金低剂量组、温郁金高剂量组和氟西汀组大鼠神经功能明显受损(P<0.01);应激第14、21天温郁金高剂量组大鼠神经功能评分明显低于PSD组(P<0.05);HE染色结果显示温郁金高剂量组大鼠病理评分显著低于PSD组(P<0.01);免疫组化检测结果显示,温郁金高剂量组大鼠海马区BrdU阳性细胞数和Nestin阳性细胞数均显著高于PSD组(P<0.01)。结论温郁金可通过降低神经功能缺损评分、减轻海马区病理损伤,增加海马区BrdU和Nestin的表达,促进PSD大鼠海马区神经再生来发挥对PSD大鼠的保护作用,但其具体作用机制需进一步探索。 Objective To investigate the effects and mechanisms of Curcuma wenyujin(Wenyujin)against post-stroke depression(PSD)and provide a research basis for elucidating the connotation of"removing blood stasis and promoting regeneration"in Traditional Chinese Medicine.Methods A rat model of PSD was established by middle cerebral artery occlusion(MCAO)combined with 21 days of chronic unpredictable mild stress(CUMS).Rats were divided into sham operation group,stroke group,PSD group,low-dose Curcuma wenyujin group(1.4 g crude drug/kg),high-dose Curcuma wenyujin group(2.8 g crude drug/kg),and positive drug group(fluoxetine,20 mg/kg).Neurological deficits were assessed using the Zea Longa score at time points before modeling and after stress.Behavioral scores of the rats were measured.Morphological changes in hippocampal cells were observed via HE staining.Cell proliferation and neural stem cell regeneration in the hippocampus were observed and assessed using BrdU and Nestin immunohistochemical staining.Results Compared with the sham operation group,neurological function was significantly impaired in the stroke group,PSD group,low-dose Curcuma wenyujin group,high-dose Curcuma wenyujin group,and fluoxetine group before stress and on the 7th day of stress(P<0.01).On the 14、21st day after stress,the neurological function score of the high-dose Curcuma wenyujin group was significantly lower than that of the PSD group(P<0.05).HE staining revealed significantly reduced pathological scores in the high-dose Wenyujin group compared to the PSD group(P<0.01).Immunohistochemistry demonstrated significantly higher numbers of BrdU-positive and Nestin-positive cells in the hippocampal region of the high-dose Wenyujin group than in the PSD group(P<0.01).Conclusion Wenyujin(Curcuma aromatica)exerts a protective effect on PSD rats by reducing neurological deficits,alleviating pathological damage in the hippocampal region,increasing the expression of BrdU and Nestin in the hippocampus,and promoting neural regeneration in the hippocampal area of PSD rats.However,the specific mechanism of action requires further investigation.
