摘要
该研究以东北地区代表性发酵豆制品为研究对象,采用Illumina PE250测序技术探究了其细菌菌群的多样性,并针对待研究的潜在益生菌停滞棒杆菌(Corynebacterium stationis),对其生长特点和生理特性进行了研究。结果表明:各产品中的细菌菌群具有一定相似性,但菌群丰富度有所差异。青方腐乳中代表性优势菌种包括Halanaerobium praevalensDSM-2228和嗜盐四联球菌(Tetragenococcus halophilus);红方腐乳中代表性优势菌种包括Kurthia sp.11kri321、柠檬色明串珠菌(Leuconostoc citreum)、停滞棒杆菌、假肠膜明串珠菌(Leuconostoc pseudomesenteroides)、海洋盐单胞菌(Cobetia marina)、耐冷海洋杆菌(Marinilactibacillus psychrotolerans)和嗜盐四联球菌(Tetragenococcus halophilus);豆酱中代表性优势菌种包括蛋白原酶乳杆菌(Lactobacillus rennini)和副地衣芽孢杆菌(Bacillus paralicheniformis)。形态学观察显示,停滞棒杆菌为黄色、表面光滑的杆状革兰氏阳性细菌,其生长曲线为典型的S型曲线,具有较强的耐热及耐胆盐特性,对酸度也具有一定耐受能力。该研究为发酵豆制品中功能性益生菌的筛选和利用提供了一定理论基础。
This study investigated the bacterial diversity of representative fermented soybean products from Northeast China using Illumina PE250 sequencing technology,meanwhile examined the growth and physiological properties of the potential probiotic Corynebacterium stationis.The results showed that the flora in each product had some similarities,but the flora richness was different.The representative dominant strains in grey sufu include Halaaerobium praevalens DSM-2228 and Tetragenococcus halophilus.The representative dominant strains in red sufu include Kurthia-sp.-11kri321,Leuconostoc citreum,Corynebacterium stationis,Leuconostoc pseudomesenteroides,Cobetia marina,Marinilactibacillus psychrotolerans,and Tetragenococcus halophilus.The dominant bacteria in soybean paste are mostly Lactobacillus rennini and Bacillus paralicheniformis.Corynebacterium stationis was determined to be a yellow,smooth-surfaced,rod-shaped Gram-positive bacterium.Its growth curve is a typical S-shaped curve,it also has strong heat resistance and bile salt resistance,and certain tolerance to acidity.This study provides a theoretical basis for the screening and utilization of functional probiotics in fermented soybean products.
