摘要
目的 探究Bax/Bcl-2/Caspase-3信号通路在黄芪多糖联合维生素C对镉致雌性大鼠生殖系统损伤的保护作用。方法 24只SD雌鼠随机分4组(每组6只):对照组腹腔注射0.9%氯化钠溶液,Cd组腹腔注射Cd Cl2,给药2组给予灌胃Aps+Vc(高低剂量)处理。试验结束后,检测血清E2、P4含量,观察卵巢病理及细胞凋亡,检测Bax、Bcl-2、Caspase-3蛋白表达。结果 腹腔注射Cd Cl2,经黄芪多糖联合维生素C干预后,Aps+Vc低、高剂量组体毛光泽、进食量和取水量、对外界刺激反应均有明显改善,腹部未出现硬块、坏疽等现象;体重增长率、卵巢指数相对升高。ELISA结果显示,Aps+Vc低、高剂量组E2、P4水平显著提高。HE染色结果显示,Aps+Vc低、高剂量组卵巢组织微环境得到明显改善。TUNEL结果显示Aps+Vc低、高剂量组卵巢组织细胞凋亡率显著下降。IHC结果显示,Bax和Caspase-3蛋白在Aps+Vc低、高剂量组卵巢组织细胞中表达率显著降低;Bcl-2蛋白在卵巢组织细胞中表达率显著升高。Western blot结果显示,Aps+Vc低、高剂量组卵巢组织细胞中Bax和Caspase-3蛋白相对表达量下调、Bcl-2蛋白相对表达量上调,Bax/Bcl-2比值降低。结论 黄芪多糖联合维生素C可通过调控镉染毒雌性大鼠卵巢组织细胞中Bax/Bcl-2、Caspase-3蛋白的表达抑制镉促进卵巢组织细胞的凋亡,调整卵巢组织细胞的凋亡过程,有效减少闭锁卵泡的生成,恢复雌性大鼠血清雌性激素水平,减少镉对卵巢的损伤,改善卵巢微环境并恢复卵巢功能,对临床上卵巢受到镉损伤后的治疗具有重要价值。
Objective To investigate the protective effect of Astragalus polysaccharides(APS)combined with vitamin C(Vc)on cadmium-induced reproductive system damage in female rats by regulating the B cell lymphoma-2(Bcl-2)-associated X protein(Bax)/Bcl-2/Caspase-3 signaling pathway.Methods 24 SDfemale mice were randomly divided into 4 groups(6 mice in each group):the control group received intraperitoneal injection of physiological saline,the Cd group received intraperitoneal injection of CdCl2,and the two treatment groups were given oral administration of Aps+Vc(high and low doses).After the experiment,serum E2 and P4 levels were measured,ovarian pathology and cell apoptosis were observed,and Bax,Bcl-2,and Caspase-3 protein expression were detected.Results Rats in the low-dose and high-dose APS+Vc groups had improved body hair gloss,food and water intake,response to external stimuli,and increased weight growth rate and ovary index.There were no signs of hard lumps or gangrene in the abdomen.ELISA results showed significant increases in serum E2 and P4 in the low-dose and high-dose APS+Vc groups.H&E staining visualized a significantly improvement in the ovarian tissue microenvironment of the low-dose and high-dose APS+Vc groups.TUNEL assay revealed a significantly decreased apoptotic rate in rat ovarian tissue of the low-dose and high-dose APS+Vc groups.IHC results showed significant decreases in positive expression of Bax and Caspase-3,and increases in Bcl-2 in the low-dose and high-dose APS+Vc groups.Western blot revealed a significantly downregulated Bax and Caspase-3,upregulated Bcl-2,and decreased Bax/Bcl-2 in rat ovarian tissue of the low-dose and high-dose APS+Vc groups.Conclusion APS combined with Vc can inhibit cadmium-induced apoptosis in ovarian tissue of female rats by regulating the protein expressions of Bax/Bcl-2 and Caspase-3,adjusting the apoptosis of ovarian tissue,reducing the generation of atresia follicles,restoring serum estrogen,alleviating ovarian damages,improving ovarian microenvironment and ovarian function.The treatment of APS combined with Vc against cadmium-induced ovarian damage in clinical practice is of great value.It has important value in clinical treatment.
