针灸“扶正固肾”穴方对顺铂小鼠肾脏组织中p-JNK、p-ERK和p-p38的影响

The effects of “strengthening healthy qi and consolidating kidney” acupuncture-moxibustion acupoint prescription on p-JNK,p-ERK and p-p38 in the kidney tissue of DDP-induced mice

摘要 目的 观察“扶正固肾”针灸穴方对顺铂(DDP)肾损伤小鼠肾脏组织中p-JNK、p-ERK和p-p38的影响,探究针灸改善化疗致肾损伤的作用机制。方法 将40只小鼠随机划分为4组,即空白组、模型组、针刺组与艾灸组,每组各10只。除空白组外,其余3组小鼠均采用腹腔注射10 mg/kg顺铂(DDP)的方式进行造模,造模过程在24 h内完成;空白组则腹腔注射等量的0.9%氯化钠溶液。针刺组与艾灸组均选取“大椎”“肝俞”“肾俞”“足三里”,分别进行针刺与艾灸治疗,其余两组仅陪同固定,1次/d,连续5 d。禁食1 d后取材,通过HE染色法对小鼠肾脏组织的病理改变进行观察,ELISA法和Western Blot法检测肾脏组织中磷酸化c-Jun氨基末端激酶(p-JNK)、磷酸化细胞外信号调节蛋白激酶(p-ERK)和磷酸化丝裂原活化蛋白激酶(p-p38)的含量和蛋白表达。结果 相比于空白组,模型组肾脏组织中p-JNK、p-ERK和p-p38含量、蛋白表达均增多(P<0.05);相比于模型组,针刺组和艾灸组肾脏组织中p-JNK、p-ERK和p-p38含量、蛋白表达均减少(P<0.05)。结论“扶正固肾”针灸能够改善化疗所致肾损伤,其作用机制或许与下调JNK、ERK、p38的磷酸化水平存在关联。 Objective To observe the effects of“strengthening healthy qi and consolidating kidney”acupuncture-moxibustion acupoint prescription on p-JNK,p-ERK and p-p38 in kidney tissue of mice with cisplatin(DDP)kidney injury,and to explore the mechanism of acupuncture and moxibustion to improve chemotherapy-induced kidney injury.Methods Forty clean male Kunming mice were divided into four groups(n=10 each):the blank,model,acupuncture,and moxibustion groups.Except blank group,the other three groups were modeled by intraperitoneal injection of DDP(10 mg/kg).The modeling was completed within 24 hours,and the blank group was injected with the same dose of 0.9%sodium chloride solution.“Dazhui(GV 14)”,“Ganshu(BL 18)”,“Shenshu(BL 23)”and“Zusanli(ST 36)”were selected in acupuncture group and moxibustion group,and acupuncture and moxibustion were administered respectively,and the other two groups were only restrained without treatment once a day for 5 consecutive days.After fasting for 1 day,samples were collected.Hematoxylin-eosin(HE)staining was used to observe pathological changes in the mouse kidney tissues.ELISA and Western blot(WB)measured contents and protein expression of phosphorylated c-Jun amino-terminal kinase(p-JNK),phosphorylated extracellular signal-regulated protein kinase(p-ERK)and phosphorylated p38 mitogen-activated protein kinase(p-p38)in renal tissues.Results Compared with the blank group,contents and protein expressions of p-JNK,p-ERK and p-p38 in kidney tissues of model group were increased(P<0.05).Compared with model group,the contents and protein expressions of p-JNK,p-ERK and p-p38 in kidney tissues of acupuncture group and moxibustion group were decreased(P<0.05).Conclusion Acupuncture and moxibustion can improve kidney injury induced by DDP,and its mechanism may be related to downregulating JNK,ERK and p38 phosphorylation levels.
