摘要
为建立叶用莴苣高效稳定的再生体系,本研究分别以叶用莴苣不同品种为外植体材料,探究不同基因型和不同浓度的激素配比组合对叶用莴苣愈伤诱导、不定芽分化和植株再生的影响。结果表明:大速生、罗马、优湖801的子叶和下胚轴的最佳再生培养基均为MS+0.3 mg/L IAA+0.5 mg/L KT。三种不同基因型叶用莴苣的子叶与下胚轴在相同培养基条件下,出愈率和出芽率均达到较高水平(出愈率95%~100%,出芽率65%~100%)。当IAA浓度为0.3 mg/L时,生根效果最好。本研究结果可为该作物的高效组织培养和品种改良提供参考。
To establish an efficient and stable regeneration system for leaf lettuce,this study used different leaf lettuce cultivars as explants to investigate the effects of varying genetic backgrounds and hormone concentration combinations on callus induction,adventitious bud differentiation,and plant regeneration.Results indicated that the optimal regeneration medium for cotyledons and hypocotyls of Da Susheng,Roma,and Youhu 801 was MS+0.3 mg/L IAA+0.5 mg/L KT.Under identical medium conditions,both cotyledons and hypocotyls from the three leaf lettuce genotypes achieved high callus formation rates(95%~100%)and budding rates(65%~100%).Root formation was most effective at an IAA concentration of 0.3 mg/L.These findings provide valuable references for efficient tissue culture and variety improvement in this crop.
