基于PI3K/Akt/FoxO3a信号通路探讨针刀改善制动诱导大鼠腓肠肌萎缩的作用机制

摘要 目的:通过PI3K/Akt/FoxO3a通路探讨针刀干预对制动诱导的大鼠腓肠肌萎缩的作用机制。方法:将32只Wistar雄性大鼠随机分为空白组、模型组、牵伸组和针刀组,采用右后肢踝关节跖屈位固定4周来制备模型。造模成功后,牵伸组沿着大鼠右踝关节背屈方向垂直于足底区方向施加0.3 N的力进行牵伸。针刀组选用直径0.4 mm×25 mm针刀,在肌肉表面筋膜处松解后,进行牵伸,牵伸方法同牵伸组。干预后测量踝关节背屈活动度ROM;检测腓肠肌肌力、腓肠肌湿重及长度;RT-PCR法检测腓肠肌PI3K、Akt、FoxO3a、MyoD1及Pax7 mRNA的表达;Western blot检测PI3K、Akt、p-PI3K及p-Akt的表达。结果:针刀组踝关节背屈ROM、腓肠肌肌力、肌肉湿重及长度、PI3K、Akt、MyoD1及Pax7 mRNA表达水平、p-PI3K和p-Akt蛋白表达水平较模型组和牵伸组均增加,差异具有统计学意义(P<0.05或P<0.01),FoxO3a mRNA表达水平降低,差异具有统计学意义(P<0.01)。结论:针刀可有效延缓大鼠腓肠肌萎缩进程,其机制可能与PI3K/Akt/FoxO3a信号通路密切相关。 Objective:To investigate the mechanism by which acupotomy intervenes in immobilization-induced gastrocnemius muscle atrophy in rats via the PI3K/Akt/FoxO3a signaling pathway.Methods:32 male Wistar rats were randomly assigned to the blank control,the model group,the stretching group,and the acupotomy group.The immobilization-induced gastrocnemius muscle atrophy model was established by fixing the right hindlimb ankle joint in plantar flexion for 4 weeks.After successful modeling,the stretching group received a 0.3 N force applied along the dorsiflexion direction of the right ankle,perpendicular to the plantar surface.In the acupotomy group,a 0.4 mm×25 mm acupotomy needle was used to release the fascia on the muscle surface,followed by the same stretching method as the stretching group.Following the intervention,ankle dorsiflexion range of motion(ROM)was measured,and gastrocnemius muscle strength,wet weight,and length were assessed.RT-PCR was used to measure mRNA expressions of PI3K,Akt,FoxO3a,MyoD1,and Pax7 in the gastrocnemius,while Western blot was performed to detect the protein expressions of PI3K,Akt,p-PI3K,and p-Akt.Results:In the acupotomy group,ankle dorsiflexion ROM,gastrocnemius muscle strength,wet weight,and length,mRNA expressions of PI3K,Akt,MyoD1,and Pax7,as well as protein expressions of p-PI3K and p-Akt were all significantly increased(P<0.05,P<0.01),and FoxO3a mRNA expression was significantly decreased compared with the model group and stretching group after the treatment(P<0.01).Conclusion:Acupotomy can effectively delay gastrocnemius muscle atrophy in rats,and its mechanism may be closely related to the PI3K/Akt/FoxO3a signaling pathway.
