基于MALDI-TOF MS的核酸质谱技术在兽医检测领域中的应用研究进展

摘要 基于基质辅助激光解析电离飞行时间质谱(matrix-assisted laser desorption/ionization time-of-fl ight mass spectrometry,MALDI-TOF MS)的核酸质谱技术是以核酸特征谱图分析为基础的快速检测技术,可实现对动物病原体核酸的高通量、高灵敏度检测。近年来,该技术因具有敏感性高、特异性强以及高通量等优势,在兽医检测领域中显现出应用潜力。本文阐述了该技术的基本原理及其在病原高通量检测、基因分型与耐药性检测方面的应用进展,提出了应用中存在的局限性及未来发展方向,为该技术的进一步应用提供参考依据。 Animal pathogen nucleic acids could be detecte d by matrix-assisted laser desorption/ionization time-offlight mass spectrometry(MALDI-TOF MS)with high-throughput and high-sensitivity that was developed based on nucleic acid characteristic spectra analysis.In recent years,MALDI-TOF MS had been promising in the fi eld of veterinary detection,benefi ting from its capabilities of high sensitivity,strong specifi city and high-throughput.Here,its fundamental principles and advances in pathogen detection with high-throughput,genotyping and antimicrobial resistance detection were described,its limitations in practice and way forward were introduced,aiming to guide its future application as a reference.
