摘要
目的对青海湖地区国家一级保护动物普氏原羚(Procapra przewalskii)中隐孢子虫(Cryptosporidium)的感染流行情况展开调查。方法基于隐孢子虫18S rRNA基因的巢式PCR方法对采集到的普氏原羚新鲜粪便样品基因组DNA进行扩增,以检测隐孢子虫感染率,并基于gp60基因片段对隐孢子虫阳性样本进行亚型分析。结果普氏原羚中隐孢子虫的感染率为3.67%(16/436),共检测到肖氏隐孢子虫(C.xiaoi)和泛在隐孢子虫(C.ubiquitum)2种隐孢子虫,其中C.xiaoi为优势虫株,占阳性样品的68.75%(11/16)。对C.ubiquitum阳性样本进行亚型分析,鉴定出其为XIIa型。结论本次研究首次证实普氏原羚中存在隐孢子虫感染,并且存在能够感染人的XIIa亚型C.ubiquitum,具有较大的人兽共患病传播风险。
This study was aimed at determining the prevalence of Cryptosporidium spp.in Procapra przewalskii,a nationally protected first-class species in China,and evaluated the zoonotic potential of the isolates.Fresh fecal specimens(n=436)were collected from P.przewalskii in the Qinghai Lake region.Nested PCR targeting the conserved region of the Cryptosporidium 18S rRNA gene was used for primary detection.Positive samples were subtyped with the gp60 gene fragment.An overall infection rate of 3.67%(16/436)was observed.Molecular characterization identified two species:Cryptosporidium xiaoi(68.75%,11/16)and C.ubiquitum(31.25%,5/16).C.ubiquitum positive samples were subtyped as XIIa.This study provides the first confirmation of Cryptosporidium infection in P.przewalskii.The presence of C.ubiquitum subtype XIIa,which can infect humans,indicates high zoonotic risk.Therefore,further surveillance and management are needed to safeguard the health of both P.przewalskii and humans.
