摘要
目的:探究洋甘菊黄酮在斑马鱼体内的舒缓、抗炎和控油功效。方法:建立斑马鱼胚胎刺激模型和炎症模型,通过分析斑马鱼的自旋次数、卵黄囊面积、心跳次数、活性氧(ROS)相对生成量及炎症因子含量,探究洋甘菊黄酮的舒缓抗炎功效;测定斑马鱼体内5α-还原酶相关基因的表达水平评估其控油功效。结果:斑马鱼胚胎在十二烷基硫酸钠(SDS)的刺激下,胚胎的自旋次数显著增加,经不同浓度洋甘菊黄酮溶液孵育后,斑马鱼胚胎自旋次数出现了不同程度的减少。洋甘菊黄酮可以降低脂多糖(LPS)造成的斑马鱼全身炎症,减少卵黄囊面积、降低心跳次数、降低ROS相对生成量、降低促炎因子白细胞介素-6(IL-6)以及肿瘤坏死因子-α(TNF-α)的含量。洋甘菊黄酮对5α-还原酶合成的关键基因srd5a1和srd5a2的相对表达量具有显著的抑制效果。结论:通过斑马鱼模型,验证了洋甘菊黄酮具有显著的舒缓、抗炎和控油功效,可为洋甘菊黄酮作为化妆品原料应用提供理论依据。
Objective:To investigate the soothing,anti-inflammatory,and oil-controlling effects of chamomile flavonoids in zebrafish.Methods:Zebrafish embryo stimulation models and inflammation models were established.The soothing and anti-inflammatory effects of chamomile flavonoids were explored by analyzing the spinning frequency,yolk sac area,heart rate,relative reactive oxygen species(ROS)production,and inflammatory factor levels of zebrafish.The oil-controlling efficacy was evaluated by determining the expression levels of 5α-reductase-related genes in zebrafish.Results:Under stimulation with sodium dodecyl sulfate(SDS),the spinning frequency of zebrafish embryos increased significantly.After incubation with chamomile flavonoid solutions of different concentrations,the spinning frequency of zebrafish embryos decreased to varying degrees.Chamomile flavonoids could alleviate systemic inflammation induced by lipopolysaccharide(LPS)in zebrafish,as evidenced by reduced yolk sac area,decreased heart rate,lowered relative ROS production,and reduced levels of the pro-inflammatory cytokines interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α).Additionally,chamomile flavonoids exhibited a significant inhibitory effect on the relative expression levels of the srd5a1 and srd5a2 genes.Conclusion:Using zebrafish models,this study verified that chamomile flavonoids possess significant soothing,anti-inflammatory,and oil-controlling effects.These findings provided a theoretical basis for the application of chamomile flavonoids as a cosmetic ingredient.
