葛根素通过调节miR-506-5p/Notch1/Hes-1信号通路抑制宫颈癌细胞增殖、迁移和侵袭

Puerarin inhibits cervical cancer cell proliferation,migration and invasion by regulating the miR-506-5p/Notch1/Hes-1 signaling pathway

摘要 目的 基于微小RNA(miRNA)506-5p/Notch1/Hes-1信号通路探究葛根素对宫颈癌细胞增殖、迁移和侵袭行为的影响。方法 使用不同浓度葛根素处理人宫颈癌细胞系HeLa,依据葛根素浓度将细胞分为:对照组(0μmol/L)、低浓度组(5μmol/L)、中浓度组(20μmol/L)和高浓度组(50μmol/L)。通过CCK-8法、划痕实验、Transwell实验分别检测细胞增殖、迁移和侵袭能力,Western blot检测促凋亡蛋白Bax、Bad和Notch信号通路蛋白Notch1、Hes-1表达水平,qRT-PCR检测miR-506-5p表达水平,细胞免疫荧光染色检测细胞周期蛋白Cyclin D1和凋亡蛋白Bax表达水平。使用miR-506-5p表达干扰质粒转染HeLa细胞,并用20μmol/L葛根素处理,Western blot检测Bax、Bad、Notch1、Hes-1和Cyclin D1蛋白表达水平。结果 与对照组相比,5~50μmol/L葛根素处理均能显著抑制HeLa细胞的增殖、迁移和侵袭活性(均P<0.05)。与对照组相比,葛根素处理组细胞Bax、Bad、miR-506-5p表达水平升高(P<0.05),而Cyclin D1、Notch1和Hes-1蛋白表达水平降低。miR-506-5p表达敲低后,Bax、Bad表达水平显著降低,Notch1、Hes-1和Cyclin D1表达水平显著升高(P<0.05)。结论 葛根素通过调节miR-506-5p表达,抑制Notch1/Hes-1通路表达,进而抑制宫颈癌细胞增殖、迁移和侵袭。 Objective To investigate the effect of puerarin on the proliferation,migration and invasion of cervical cancer cells based on the microRNA(miRNA)506-5p/Notch1/Hes-1 signaling pathway.Methods Human cervical cancer cell line HeLa was treated with different concentrations of puerarin and divided into control group(0μmol/L),low concentration group(5μmol/L),medium concentration group(20μmol/L)and high concentration group(50μmol/L)according to the concentration of puerarin.Cell proliferation,migration and invasion ability were detected by CCK-8,scratch assay and Transwell assay,respectively.The expression levels of pro-apoptotic proteins Bax and Bad and Notch signaling pathway proteins Notch1 and Hes-1 were detected by Western blot,the expression level of miR-506-5p was detected by qRT-PCR,and the expression levels of cell cycle protein Cyclin D1 and apoptotic protein Bax were detected by cell immunofluorescence staining.The expression vector of miR-506-5p was transfected into HeLa cells and treated with 20μmol/L puerarin,and the protein expression levels of Bax,Bad,Notch1,Hes-1 and Cyclin D1 were detected by Western blot.Results Compared with the control group,puerarin at 5-50μmol/L significantly inhibited the proliferation,migration and invasion of HeLa cells(all P<0.05).Compared with the control group,the Bax,Bad,miR-506-5p expression levels were increased(P<0.05),and the Cyclin D1,Notch1 and Hes-1 protein expression levels were decreased in puerarin treatment group.After the expression of miR-506-5p was knocked down,the expression levels of Bax and Bad were significantly decreased,and the expression levels of Notch1,Hes-1 and Cyclin D1 were significantly increased(P<0.05).Conclusion Puerarin inhibits the proliferation,migration and invasion of cervical cancer cells by regulating the expression of miR-506-5p and inhibiting the expression of Notch1/Hes-1 pathway.
