摘要
目的 探讨孕期六氟环氧丙烷二聚羧酸(HFPO-DA)暴露对母鼠及其胎鼠肝脏损伤的影响。方法 建立孕期大鼠HFPO-DA暴露模型,将孕0.5 d的SD大鼠随机分为空白对照组(给予去离子水)和不同浓度HFPO-DA暴露组(分别给予1 mg/kg、10 mg/kg、100 mg/kg的HFPO-DA)。实验组通过灌胃暴露至孕19.5 d,随后处死孕鼠。记录孕鼠及胎鼠的体质量和肝脏质量,采用苏木精–伊红染色(H-E染色)观察肝脏组织病理改变;全自动生化分析仪用于测定孕鼠血清中天冬氨酸氨基转移酶(AST)、碱性磷酸酶(ALP)、丙氨酸氨基转移酶(ALT)、总胆固醇(THO)、高密度脂蛋白胆固醇(HDL-C)水平、白蛋白(ALB)、总甘油三酯(TG)、总胆汁酸(TBA)和低密度脂蛋白胆固醇(LDL-C)。结果 与对照组相比,高剂量HFPO-DA暴露组孕鼠的肝脏质量和肝脏系数显著增加。H-E组织染色显示该组孕鼠肝脏汇管区充血、肝细胞点状坏死及肝细胞水肿。高剂量HFPO-DA组孕鼠血清中ALT、TBA、LDL-C水平显著高于对照组;此外,中剂量和高剂量组的HDL-C水平均显著低于对照组。胎鼠暴露组与对照组相比,肝脏质量及病理组织未见明显异常。结论 孕期HFPO-DA暴露可导致SD大鼠母鼠肝脏损伤及代谢指标异常,但对胎鼠的影响较小。
Objective To investigate the effects of hexafluoropropylene oxide dimer acid(HFPO-DA)exposure during pregnancy on liver damage in maternal rats and their fetuses.Methods A rat model of HFPO-DA exposure during pregnancy was established.The pregnant Sprague-Dawley(SD)rats were randomly allocated to four groups on day 0.5 of gestation:A control group(administered deionized water)and three HFPO-DA exposure groups(receiving HFPO-DA via gavage at doses of 1 mg/kg,10 mg/kg,and 100 mg/kg).All treatment groups underwent continuous gavage administration until day 19.5 of gestation,after which the pregnant rats were euthanized.Maternal and fetal body weights and liver weights were taken.Hematoxylin and eosin(H-E)staining was used to observe liver histopathological changes.Aspartate transferase(AST),alkaline phosphatase(ALP),Alanine amino–transferase(ALT),total cholesterol(THO),high-density lipoprotein cholesterol(HDL-C),albumin(ALB),triglycerides(TG),total bile acids(TBA),low-density lipoprotein cholesterol(LDL-C)were measured using an automated biochemical analyzer.Results Compared to the control group,the high-dose HFPO-DA group showed a significant increase in liver weight and organ coefficient in maternal rats.H-E tissue staining revealed congestion in the periportal areas,focal necrosis of hepatocytes,and hepatocellular edema in the high-dose group.The levels of ALT,TBA,and LDL-C in serum were significantly higher in the high-dose HFPO-DA group compared to the control group.Furthermore,HDL-C levels in the middle and high-dose groups were significantly lower than control group.No significant changes were observed in the liver weight or histopathological changes in the fetal rats exposed to HFPO-DA.Conclusion Exposure to HFPO-DA during pregnancy can cause liver damage and metabolic abnormalities in maternal rats,but has a relatively small effect on fetal rats.
