摘要
建立高效液相色谱方法测定氟苯尼考产品中氟苯尼考对映异构体的含量。采用直链淀粉-三(3,5-二甲基苯基氨基甲酸酯)键合硅胶为填充柱(ChiralpakAD-H,5um,4.6mm×250mm),以正己烷-异丙醇-甲醇(700:150:150,VV)为流动相,流速为1.0mL/min,柱温为30℃,检测波长为224nm。结果表明:氟苯尼考对映异构体在0.04~4.00ug/mL内线性关系良好,平均回收率为98.3%~99.8% RSD为0.2%~6.4%。本方法专属性强、准确度高、分析时间短,可用于氟苯尼考产品中氟苯尼考对映异构体的质量控制。
A HPLC method was established for the determination of the enantiomer of florfenicol.A Daicel Chiralpak AD-H column(5μm,4.6 mm×250 mm)was used,with the mobile phase of n-hexane-isopropyl alcohol-methanol(700:150:150,V/V);detection wavelength was 224 nm;flow rate was 1.0 mL/min.Calibration curve was good linear in the range of 0.04~4.00μg/mL and average recovery was 98.3%~99.8%,with RSD of 0.2%~6.4%.This method has strong specificity,high accuracy,and short analysis time.It can be used for quality control of enantiomer in florfenicol.
