光污染条件下的水稻抽穗期QTL定位分析及其候选基因鉴定

QTL mapping analysis and candidate genes identification of rice heading date under light pollution conditions

摘要 随着我国城镇化程度的不断提高,光污染对植物尤其是水稻等作物的影响将越来越大,对光污染相关基因进行深入研究意义重大.本研究旨在光污染条件下对水稻抽穗期进行QTL定位分析,并发掘光污染条件下的抽穗期基因,为水稻抗光污染分子育种提供科学依据.利用光敏感粳稻轮回亲本Sasanishiki、光钝感籼稻供体品种Habataki,以及它们杂交衍生的一套染色体单片段代换系(CSSL),将它们分期播种,分别种植在正常条件和光污染条件下,调查抽穗期并进行QTL定位分析.结果在正常条件下检测到1个位于8号染色体的qDTH8,加性效应值为3.5,是个新位点;在光污染条件下检测到4个QTLs:qDTH1,qDTH2,qDTH3-1和qDTH3-2,前三个QTLs的加性效应值分别为3,2.5和2.5,qDTH3-2的加性效应值范围为-5~-4.仅有qDTH3-2在两个播期的光污染条件下被重复检测到,说明该QTL在长光照下稳定表达.进一步将qDTH3-2精细定位在SNP标记m122和m137之间19.1 kb的区段上.基因预测发现该区段内有3个候选基因:LOC_Os03g54070,LOC_Os03g54084和LOC_Os03g54091,其中LOC_Os03g54084是光敏色素C(phytochrome C,phyC),编码组氨酸激酶,推测该基因是qDTH3-2的候选基因.综上,在正常条件和光污染条件下分别重复检测到1个抽穗期QTL:qDTH8和qDTH3-2,进一步将qDTH3-2精细定位在19.1 kb的区段上,基因预测表明该基因是光敏色素C,为光污染下水稻抽穗期育种提供新的选择目标. With the continuous improvement of urbanization in China,the impact of light pollution on plants,especially rice and other crops,will become increasingly significant.It is of great significance to conduct in-depth research on light pollution related genes.This study aimed to conduct QTL mapping analysis for heading date of rice under light pollution conditions,and to provide a theoretical basis for exploring heading date genes under light pollution conditions.In this study,we used the japonica transmigration parent Sasanishiki,the indica donor variety Habataki,and a set of chromosome single-segment substitution lines(CSSL)derived from their hybrids.They were sown in stages under normal conditions and light pollution conditions,respectively,and the heading time was investigated,and QTL mapping analysis was conducted.Under normal conditions,a new QTL,qDTH8 with 3.5 additive effect value,was detected on chromosome 8;Four QTLs were detected under light pollution conditions,qDTH1,qDTH2,qDTH3-1,and qDTH3-2.The additive effect values of the first three QTLs were 3,2.5,and 2.5,while the additive effect value range of qDTH3-2 was−5 to−4.Only qDTH3-2 was repeatedly detected under light pollution conditions at two sowing dates,indicating stable expression of this QTL under long light conditions.Further refine the positioning of qDTH3-2 to the 19.1 kb region between SNP markers m122 and m137.Gene prediction revealed three candidate genes within this region:LOC-Os03g54070,LOC-Os03g54084,and LOCOs03g54091.Among them,LOC-Os03g54084 is a candidate gene for qDTH3-2,encoding histidine kinase(phyC),inferred and identified that this gene is a candidate gene for qDTH3-2.In summary,under normal conditions and light pollution conditions,qDTH8 and qDTH3-2 were repeatedly located at one heading date.Furthermore,qDTH3-2 was finely mapped between the 19.1 kb SNP markers.Gene prediction showed that this gene is photosensitive pigment C,providing a new selection target for rice heading to breeding under light pollution.
