一株抗马铃薯疮痂病细菌MY-01的分离鉴定及发酵条件优化

摘要 为筛选适宜马铃薯疮痂病的生防资源,进而推动马铃薯产业绿色可持续发展,从黑龙江省东南部牡丹江市温春镇马铃薯种植区来源菌株中筛选出一株抗疮痂病的菌株。通过形态学观察、生理生化特性分析及16S rDNA分子进行鉴定,并对其发酵粗提物的抑菌活性进行发酵条件优化。通过平板对峙法和琼脂扩散法从马铃薯种植区筛选出一株对疮痂链霉菌抑菌效果最好的菌株;以琼脂扩散法所得的抑菌圈直径为评判标准,利用单因素实验法、Plackett-Burman试验设计和响应面法优化其培养基和发酵条件。结果表明,对疮痂链霉菌抑菌效果最好的菌株MY-01,经鉴定为解淀粉芽孢杆菌(Bacillus amyloliquefaciens);MY-01最佳培养基配比为葡萄糖22.4 g·L^(-1),酵母浸粉5 g·L^(-1),KH_(2)PO_(4)1.9 g·L^(-1);培养时间为40 h,温度为31℃,pH为7.5,转速为180 r·min^(-1),装液量100 mL∶500 mL,优化后抑菌圈直径由18.18 mm提高到了33.84 mm。综上,B.amyloliquefaciens菌株MY-01对马铃薯疮痂病致病菌疮痂链霉菌有抑制作用。 In order to screen suitable biocontrol resources for potato scab disease and promote green and sustainable development of the potato industry.We screened a strain resistant to scab disease from the potato planting area of Wenchun Town,Mudanjiang City,in the southeastern part of Heilongjiang Province.Through morphological observation,physiological and biochemical characteristic analysis,and 16S rDNA molecular identification,the fermentation conditions of its crude extract were optimized for antibacterial activity.A strain with the best antibacterial effect against Streptomyces scabies was selected from the potato planting area using plate confrontation and agar diffusion methods.Based on the diameter of the antibacterial zone obtained by agar diffusion,single-factor experiments,Plackett-Burman design,and response surface methodology were used to optimize its culture medium and fermentation conditions.The strain MY-01,which showed the best antibacterial effect against S.scabies,was identified as Bacillu amyloliquefaciens.The optimal medium composition for MY-01 was glucose 22.4 g·L^(-1),yeast extract 5 g·L^(-1),KH_(2)PO_(4)1.9 g·L^(-1).The culture time was 40 hours,temperature 31℃,pH7.5,rotation speed 180 r·min^(-1),and liquid volume 100 mL∶500 mL.After optimization,the diameter of the antibacterial zone increased from 18.18 mm to 33.84 mm.In conclusion,B.amyloliquefaciens MY-01 has inhibitory effects on the pathogen of potato scab disease.
