能力验证饮用水菌落总数的质量控制及测量不确定度评定

Quality Control and Measurement Uncertainty Evaluation for Proficiency Testing of the Total Number of Bacteria Colonies in Drinking Water

摘要 为了提高实验室的菌落总数检测能力,保证检测结果的准确性和可靠性,本实验室参加爱德士公司组织的菌落总数能力验证,从“人、机、料、法、环”五个方面对检测全过程进行质量控制。依据《生活饮用水标准检验方法 第12部分:微生物指标》(GB/T 5750.12—2023)中4.1平皿计数法和4.2酶底物法的要求,对能力验证盲样进行检测,能力验证结果为满意。同时,对平皿计数法和酶底物法检测饮用水菌落总数的测量不确定度进行评定,通过重复性测定、分度吸量管取样和数据修约对不确定度来源进行分析。当包含概率p=95%,包含因子k=2.26时,平皿计数法和酶底物法检测到能力验证盲样中菌落总数的扩展不确定度分别为0.099 9、0.026 2(以对数计)。取反对数,得平皿计数法和酶底物法检测能力验证盲样中菌落总数的取值区间分别为121~192 CFU/mL和167~188 MPN/mL。 In order to improve the detection ability of the total number of bacteria colonies in the laboratory and ensure the accuracy and reliability of the test results,the laboratory took part in the proficiency testing of the total number of bacteria colonies organized by IDEXX,and the quality control of the entire experiment was carried out from the five aspects of“personnel,equipment,materials,methods,and environment”.The proficiency test blind samples were tested according to the requirements of Standard Examination Methods for Drinking Water-Part 12:Microbiological Parameters(GB/T 5750.12-2023)Section 4.1(Plate Counting Method)and Section 4.2(enzyme substrate method),and the results were satisfactory.The measurement uncertainty of detecting the total number of bacteria colonies in drinking water by plate counting method and enzyme substrate method was evaluated,and the sources of uncertainty were analyzed through repeatability testing,graduated pipette sampling and data rounding.On condition that the coverage pobability was 95%,and the coverage factor was 2.26,the expanded uncertainties of total number of bacteria colonies in proficiency test blind samples respectively were 0.0999 and 0.0262(log scale)for the plate counting method and enzyme substrate method.After taking the anti-logarithm,the total number of bacteria colonies in proficiency test blind samples by plate counting method and enzyme substrate method respectively were 121-192 CFU/mL,and 167-188 MPN/mL.
