摘要
目的建立高效液相色谱法(high performance liquid chromatography,HPLC)检测动物软骨提取物中硫酸软骨素含量的分析方法。方法样品经乙腈提取,采用WatersAtlantis T3色谱柱(4.6 mm×250 mm,5μm)进行分离,对流动相的组成和洗脱方式等参数进行优化和探讨。结果以乙腈-戊烷磺酸钠溶液为流动相,等度洗脱,柱温35℃,可以使硫酸软骨素10 min内实现基线分离。在192 nm的检测波长下,在5~200µg/mL的质量浓度范围内呈现良好的线性关系,相关系数大于0.999。硫酸软骨素在5、50和100 mg/g添加水平的回收率为85.6%~103.5%,相对标准偏差小于3%,方法检出限为1.0 mg/g、定量限为3.0 mg/g。结论该方法操作简便、分析快速、结果准确且灵敏度优异,适用于动物软骨提取物中硫酸软骨素的快速检测。
Objective To establish a analytical method for determination the chondroitin sulfate in animal cartilage extracts by high performance liquid chromatography(HPLC).Methods The samples were extracted with acetonitrile and separated using a Waters Atlantis T3 column(4.6 mm×250 mm,5μm).The parameters such as the composition of the mobile phase and the elution mode were optimized and discussed.Results Using acetonitrile-sodium pentane sulfonate solution as the mobile phase,the column temperature was 35℃,and the baseline separation of chondroitin sulfate could be achieved within 10 minutes.At the detection wavelength of 192 nm,there was a good linear relationship in the mass concentration range of 5-200 mg/L with the correlation coefficient of more than 0.999.For deoxynivalenol residues at spiked concentrations of 5,50 and 100 mg/g,recovery rates varied between 85.6% and 103.5%.The relative standard deviation was less than 3%.The limits of detection and the limits of quantitation for chondroitin sulfate were 1.0 mg/g and 3.0 mg/g.Conclusion This method is simple to operate,rapid in analysis,accurate in results and has excellent sensitivity.It is suitable for the rapid detection of chondroitin sulfate in animal cartilage extracts.
