副干酪乳杆菌CCFM1367发酵葛根素缓解酒精性肝损伤的机制

摘要 目的研究副干酪乳杆菌CCFM1367(Lactobacillus paracasei CCFM1367)发酵葛根素对酒精性肝损伤的保护作用及其机制。方法将32只6周龄C57BL/6J,雄性小鼠随机分为对照组、模型组和实验组(分别接受葛根素和副干酪乳杆菌CCFM1367发酵葛根素干预),使用Lieber-DeCarli酒精液体饲料喂养5周形成慢性酒精性肝病(alcoholic liver disease,ALD)模型,检测肝脏组织病理学、血清和肝功能生化指标、实时荧光定量PCR检测基因表达、免疫荧光染色观察肠道紧密连接蛋白表达。结果与未发酵葛根素组相比,副干酪乳杆菌CCFM1367发酵葛根素明显改善了ALD小鼠肝脏组织病理损伤,调节血脂水平,降低血清脂多糖(lipopolysaccharide,LPS)水平,抑制肝脏氧化应激和炎症反应,调节酒精代谢相关基因的表达,修复酒精诱导的肠道屏障功能损伤。结论副干酪乳杆菌CCFM1367发酵葛根素可以通过缓解肝脏损伤和增强小鼠肠道屏障完整性有效缓解小鼠慢性ALD。 Aim To investigate the protective effects and underlying mechanisms of puerarin fermented by Lactobacillus paracasei CCFM1367 in alcoholic liver injury.Methods Thirty-two 6-week-old male C57BL/6J mice were randomly divided into the control group,model group,puerarin intervention group,and Lactobacillus paracasei CCFM1367-fermented puerarin intervention group.Chronic alcoholic liver disease(ALD)was induced by administering a Lieber-DeCarli alcohol liquid diet for five weeks.Assessments included histopathological examination of liver tissue,determination of serum and liver biochemical markers,analysis of gene expression via real-time quantitative PCR,and assessment of intestinal tight junction protein expression via immunofluorescence staining in the gut.Results Compared to the unfermented puerarin group,the Lactobacillus paracasei CCFM1367-fermented puerarin group showed marked improvement in liver tissue pathology,regulation of blood lipid levels,reduction of serum lipopolysaccharide(LPS)concentrations,suppression of hepatic oxidative stress and inflammation,modulation of alcohol metabolism-related gene expression and restoration of alcohol-induced intestinal barrier dysfunction.Conclusion Lactobacillus paracasei CCFM1367-fermented puerarin can effectively alleviate chronic ALD in mice by ameliorating liver injury and enhancing intestinal barrier integrity.
