摘要
为了建立‘和谐2号’毛白杨(Populus tomentosa)高效组织培养快繁体系,以茎段为外植体,通过培养基筛选和正交试验设计,构建了包括启动、增殖和生根培养的完整快繁体系。研究结果表明:①MS培养基对‘和谐2号’杨启动培养效果最佳,诱导率达80.56%;②增殖培养中,6-苄氨基嘌呤(6-BA)影响最显著,最佳培养基配方为“MS培养基+6-BA 0.8 mg/L+萘乙酸(NAA)0.3 mg/L+蔗糖20 g/L”,芽诱导率87.65%,45 d增殖倍数达3.50;③“1/2MS培养基+吲哚丁酸(IBA)0.5 mg/L+蔗糖20 g/L”为最佳生根培养基,生根率85.90%。
To establish an efficient tissue culture and rapid propagation system for Populus tomentosa‘Hexie 2’,stem segments were used as explants.A complete rapid propagation system including initiation,proliferation,and rooting culture was constructed through medium screening and orthogonal experimental design.The results showed that:(1)MS medium was the most effective for initiation culture of‘Hexie 2’P.tomentosa,with an induction rate of 80.56%.(2)In proliferation culture,6-benzylaminopurine(6-BA)had the most significant effect.The optimal medium formula was“MS medium+6-BA 0.8 mg/L+naphthaleneacetic acid(NAA)0.3 mg/L+sucrose 20 g/L”,with a bud induction rate of 87.65%and a proliferation multiple of 3.50 within 45 days.(3)“1/2 MS medium+indolebutyric acid(IBA)0.5 mg/L+sucrose 20 g/L”was the optimal rooting medium,with a rooting rate of 85.90%.
