超声辅助酶解法提取兰州大尾羊尾脂的工艺优化

Process optimization of ultrasound-assisted enzymatic hydrolysis for the extraction of Lanzhou fat-tailed sheep tail fat

摘要 为了进一步开发兰州大尾羊的经济价值,以兰州大尾羊尾巴为原料,采用超声辅助酶解法提取羊尾脂。以羊尾脂得率为评价指标,利用单因素试验筛选试验的中心点,Plackett-Burman试验筛选对羊尾脂得率有显著影响的因素,再通过响应面试验对兰州大尾羊尾脂的提取工艺进行优化,并对其理化指标和脂肪酸组成进行测定。结果表明:兰州大尾羊尾脂的最佳提取工艺为料液比1∶1.8、中性蛋白酶添加量1.0%(以羊尾样品质量计)、超声功率200 W、超声时间44.5 min、酶解时间2 h、超声酶解温度46℃,在此条件下羊尾脂得率为80.22%;提取的羊尾脂皂化值(KOH)为196.22 mg/g,碘值(I)为48.65 g/100 g,酸值(KOH)为0.92 mg/g,过氧化值为0.18 g/100 g,酸值和过氧化值均符合GB 10146—2015的要求;羊尾脂中饱和脂肪酸含量为54.035%,不饱和脂肪酸含量为45.814%。综上,可以采用超声辅助酶解法高效提取羊尾脂。 In order to further develop the economic value of Lanzhou fat-tailed sheep,Lanzhou fat-tailed sheep tail was used as the raw material and the fat was extracted by ultrasound-assisted enzymatic hydrolysis method.With the yield of the sheep tail fat as the evaluation index,the central points of the test was screened by using the single factor test,and the factors that had a significant effect on the yield of the sheep tail fat were screened by Plackett-Burman test,and then the extraction process of the sheep tail fat was optimized by response surface test.The physicochemical indexes and fatty acid composition of the sheep tail fat were determined.The results showed that the optimal extraction process of the sheep tail fat was as follows:material-liquid ratio 1∶1.8,neutral protease dosage 1.0%(based on the mass of the sheep tail),ultrasonic power 200 W,ultrasonic time 44.5 min,enzymatic hydrolysis time 2 h,and ultrasonic and enzymatic hydrolysis temperature 46℃.Under the optimal conditions,the yield of the sheep tail fat was 80.22%.The saponification value of the extracted sheep tail fat was 196.22 mgKOH/g,the iodine value was 48.65 gI/100 g,the acid value was 0.92 mgKOH/g,and the peroxide value was 0.18 g/100 g.The acid value and peroxide value were in accordance with the requirements of GB 10146-2015.The content of saturated fatty acid in the sheep tail fat was 54.035%,and the content of unsaturated fatty acid was 45.814%.In conclusion,ultrasound-assisted enzymatic hydroysis method can efficiently extract the sheep tail fat.
