青砖茶水提物对代谢相关脂肪性肝病及溃疡性结肠炎的预防作用

Preventive Effects of Aqueous Qingzhuan Tea Extract on Metabolic Dysfunction-associated Fatty Liver Disease and Ulcerative Colitis

摘要 探究青砖茶(Qing Brick Tea,QBT)水提物对代谢相关脂肪性肝病(Metabolic Dysfunction-Associated Fatty Liver Disease,MAFLD)及溃疡性结肠炎(Ulcerative Colitis,UC)的预防作用。将20只8周龄雌性C57BL/6小鼠随机分为对照组(Negative Control,NC)、模型组(Model Control,MC)、低剂量组(Low Dose,LD)和高剂量组(High Dose,HD)。高脂饲料喂养联合2.5%葡聚糖硫硫酸钠(Dextran Sulfate Sodium,DSS)自由饮水建立MAFLD及UC模型,检测小鼠体重、肝重、结肠长度、肝脏生化及血脂指标、组织病理、炎症因子的表达水平。与NC组相比,MC组小鼠肝脏指数显著增加,为0.06,呈现黄色油腻感,镜下可见大量脂质空泡;结肠长度明显缩短,为4.38 cm,且结构紊乱,腺体遭受破坏严重,大量炎性细胞浸润。与MC组相比,LD组和HD组小鼠肝脏指数明显降低,均为0.05,颜色更接近NC组,无油腻感,镜下脂质空泡明显减少;结肠长度显著增长,分别为5.04和5.28 cm,腺体结构恢复正常,炎性细胞的浸润显著减少。LD组丙氨酸转氨酶(Alanine Aminotransferase,ALT)、天门冬氨酸转氨酶(Aspartate Aminotransferase,AST)、低密度脂蛋白胆固醇(Low-Density Lipoprotein Cholesterol,LDL-C)、总胆固醇(Total Cholesterol,TC)、甘油三酯(Triglycerides,TG)分别为67.58、44.36、4.57、7.61、1.46 mmol/L,HD组分别为53.83、28.05、3.44、5.87、0.68 mmol/L,均显著降低。与MC组相比,LD组和HD组小鼠促炎因子的转录水平及炎症通路相关蛋白表达均显著降低(P<0.05),表明青砖茶水提物能够有效预防MAFLD及UC,其机制可能与抑制炎症信号通路有关。 The preventive effects of aqueous Qingzhuan tea(QBT)extract on metabolic dysfunction-associated fatty liver disease(MAFLD)and ulcerative colitis(UC)were examined.Twenty eight-week-old female C57BL/6 mice were randomly divided into negative control(NC),model control(MC),low-dose(LD),and high-dose(HD)groups.The MAFLD and UC models were established by high-fat feeding combined with 2.5%dextran sulfate sodium in drinking water ad libitum.The body weight,liver weight,colon length,biochemical markers,and blood lipid parameters of the liver,and the histopathological characteristics and inflammatory factor expression of the colon and liver were determined.Compared with that of the NC group,the MC group exhibited a significantly increased liver index of 0.06.The livers of the MC group were yellow and greasy.Under the microscope,numerous lipid vacuoles were observed.In addition,the colon was remarkably shortened to 4.38 cm,with a disordered structure,severely damaged glands,and significant inflammatory cell infiltration.Compared with that of the MC group,the LD and HD groups had significantly lower liver indexes.The livers of these treatment groups exhibited a similar color to that of the NC group and were not greasy.Microscopy images showed significantly fewer lipid vacuoles.Moreover,the colons were significantly lengthened up to 5.04 and 5.28 cm,respectively.The glandular structure was restored,and inflammatory cell infiltration decreased considerably.In the LD group,the alanine aminotransferase,aspartate aminotransferase,low-density lipoprotein cholesterol,total cholesterol,and triglyceride contents were 67.58,44.36,4.57,7.61,and 1.46 mmol/L,respectively.In the HD group,they were 53.83,28.05,3.44,5.87,and 0.68 mmol/L,respectively,indicating a significant reduction.Compared with those of the MC group,the transcription levels of pro-inflammatory factors and the expression levels of proteins related to inflammatory pathways in the LD and HD mice were significantly lower(P<0.05).These findings revealed that the aqueous QBT extract can effectively prevent MAFLD and UC.The underlying mechanisms may be associated with the inhibition of inflammatory signaling pathways.
