摘要
目的:利用网络药理学及生物信息技术研究柴胡-黄芩药对治疗支气管哮喘(BA)的信号靶点和通路。方法:利用中药系统药理学数据库与分析工具(TCMSP)筛选柴胡-黄芩药对治疗BA的活性成分及核心作用靶点,运用DisGeNET、GeneCards、OMIM数据库提取BA的作用靶点;借助韦恩图,对柴胡-黄芩的作用靶点与BA的疾病靶点取交集;构建中药-活性成分-共同靶点网络,利用网络分析插件分析重要的作用靶点;构建蛋白互作(PPI)网络筛选关键蛋白;通过DAVID数据库以及微生信云平台对共同靶点实施基因本体(GO)功能及京都基因与基因组百科全书(KEGG)通路富集分析。结果:柴胡-黄芩与BA的共同靶点有76个;PPI网络涉及76个节点,646条边,筛选后获得15个核心靶点,包括P-糖蛋白(ABCB1)、基质金属蛋白酶2(MMP2)、表皮生长因子受体(EGFR)、胱天蛋白酶3(CASP3)、热休克蛋白90α家族A类成员1(HSP90AA1)、雷帕霉素机制靶蛋白(MTOR)、聚腺苷二磷酸核糖聚合酶1(PARP1)、前列腺素内过氧化物合酶2(PTGS2)、基质金属蛋白酶9(MMP9)、非受体酪氨酸激酶(SRC)、雌激素受体1(ESR1)、蛋白激酶B(Akt1)、髓过氧化物酶(MPO)、Janus激酶2(JAK2)、磷脂酰肌醇3-激酶催化亚基γ(PIK3CG)。GO功能主要涉及调控蛋白激酶的信号传导和激酶活性。KEGG通路富集气泡图则揭示其与PI3K-Akt信号通路、代谢通路以及癌症相关的蛋白聚糖通路等多个通路的关联性。结论:研究揭示了柴胡-黄芩药对治疗BA的活性成分和关键通路等,为其治疗BA的临床研究奠定了理论基础。
Objective:To investigate the signaling targets and pathways of the Chaihu-Huangqin pair in treating bronchial asthma(BA)using network pharmacology and bioinformatics.Methods:Active components and core targets of Chaihu-Huangqin for BA treatment were screened via the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP).BA-related targets were extracted from DisGeNET,GeneCards,and OMIM databases.Intersecting targets between Chaihu-Huangqin and BA were identified using Venn diagrams.A herb-active componentcommon target network was constructed and analyzed using network analysis tools.A protein-protein interaction(PPI)network was built to identify key proteins.Gene Ontology(GO)functional enrichment and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analyses were performed on common targets using the DAVID database and the Microbioinformatics cloud platform.Results:Seventy-six common targets were identified between Chaihu-Huangqin and BA.The PPI network comprised 76 nodes and 646 edges,with 15 core targets screened:ABCB1,MMP2,EGFR,CASP3,HSP90AA1,MTOR,PARP1,PTGS2,MMP9,SRC,ESR1,Akt1,MPO,JAK2,and PIK3CG.GO analysis revealed primary involvement in regulating protein kinase signal transduction and kinase activity.KEGG enrichment bubble charts indicated associations with the PI3K-Akt signaling pathway,metabolic pathways,and cancer-related proteoglycan pathways.Conclusion:This study elucidates the active components and key pathways of Chaihu-Huangqin in treating BA,providing a theoretical foundation for clinical research.
