姬松茸多糖-铬(Ⅲ)络合物的制备及降血糖活性体外评价

摘要 【目的】探究姬松茸多糖-铬(Ⅲ)络合物[Agaricus blazes polysaccharide-chromium(Ⅲ)complex,ABMP-Cr(Ⅲ)]的制备工艺、结构特性与生物活性。【方法】采用水提醇沉法从姬松茸(Agaricus blazei)中提取多糖,通过反应温度、底物浓度、反应时间和pH值的单因素及响应面法,研究ABMP-Cr(Ⅲ)的最佳制备条件;采用扫描电镜、红外光谱对其进行结构分析,通过测量α-淀粉酶和α-葡萄糖苷酶抑制率分析其降血糖活性。【结果】ABMP-Cr(Ⅲ)制备的最佳条件是反应温度22.0℃,反应时间40 min,pH值6.0,底物浓度24.00 mg·mL^(-1);在该条件下ABMP-Cr(Ⅲ)的螯合率为90.13%。姬松茸多糖(Agaricus blazei Murill polysaccharides,ABMP)呈长块状结构,ABMP-Cr(Ⅲ)表面较光滑呈片层结构;Cr^(3+)与多糖的结合位点是O-H、C-H和C-O-H。ABMP-Cr(Ⅲ)的降血糖活性随着样品浓度升高而升高,且其效果优于ABMP,当ABMP-Cr(Ⅲ)为0.6 mg·mL^(-1)时α-淀粉酶抑制率可达55.11%;当ABMP-Cr(Ⅲ)浓度为5 mg·mL^(-1)时,α-葡萄糖苷酶抑制率可达40.48%。【结论】ABMP-Cr(Ⅲ)在本研究最佳制备条件下制备效率高,其结构与ABMP差异明显,ABMP-Cr(Ⅲ)具有良好的降血糖活性,具有很好的开发前景。 【Objective】Explore the preparation process,structural characteristics,and biological activity of Agaricus blazei polysaccharide-chromium(Ⅲ)complex[ABMP-Cr(Ⅲ)].【Methods】Polysaccharides were extracted from Agaricus blazei by water extraction and alcohol precipitation.The optimum preparation conditions of ABMP-Cr(Ⅲ)were studied by single factor and response surface method of reaction temperature,substrate concentration,reaction time and pH.Its structure was analyzed by scanning electron microscopy and infrared spectroscopy,and its hypoglycemic activity was analyzed by measuring the inhibition rate ofα-amylase andα-glucosidase.【Results】The optimum conditions for the preparation of ABMP-Cr(Ⅲ)were as follows:reaction temperature 22.0°C,reaction time 40 min,pH 6.0,substrate concentration 24.00 mg·mL^(-1).Under these conditions,the chelating rate of ABMP-Cr(Ⅲ)was 90.13%.Agaricus blazei Murill polysaccharides(ABMP)had a long block structure,and the surface of ABMP-Cr(Ⅲ)was smooth and lamellar structure.The binding sites of Cr^(3+)to polysaccharides were O-H,C-H and C-O-H.The hypoglycemic activity of ABMP-Cr(Ⅲ)increased with the increase of sample concentration,and its effect was better than that of ABMP.When ABMP-Cr(Ⅲ)was 0.6 mg·mL^(-1),the inhibition rate ofα-amylase could reach 55.11%.When the concentration of ABMP-Cr(Ⅲ)was 5 mg·mL^(-1),the inhibition rate ofα-glucosidase was 40.48%.【Conclusion】ABMP-Cr(Ⅲ)has high preparation efficiency under the optimal preparation conditions in this study,and its structure is significantly different from ABMP.ABMP-Cr(Ⅲ)has good hypoglycemic activity and has good development prospects.
