人参皂苷Rh2联合顺铂对A549/DDP细胞的协同抗肿瘤效应及机制研究

Synergistic Antitumor Effect and Mechanism of Ginsenoside Rh2 and Its Combination with Cisplatin in A549/DDP Cells

摘要 目的研究人参皂苷Rh2(ginsenoside Rh2,G-Rh2)与顺铂(cisplatin,DDP)联合用药对肺癌顺铂耐药A549/DDP细胞的影响及其机制。方法采用CCK-8法检测不同浓度G-Rh2、DDP在不同时间点对A549、A549/DDP细胞的毒性作用;采用倒置显微镜观察细胞形态学变化,MDC染色和免疫荧光检测细胞自噬,Western Blot法检测耐药相关蛋白MDR1、MRP1、LRP,及细胞自噬相关蛋白p62、LC3的表达。结果G-Rh2能够显著抑制A549、A549/DDP细胞生长,且抑制作用与药物浓度和作用时间密切相关。与对照组比较,G-Rh2组和DDP组细胞生长抑制,MRP、MDR1、LRP、p62蛋白表达显著下降(P<0.01),细胞自噬水平、LC3Ⅱ/LC3I蛋白表达显著上升(P<0.01);与G-Rh2组、DDP组比较,G-Rh2+DDP组细胞生长抑制明显,MRP、MDR1、LRP、p62蛋白表达显著下降(P<0.05),细胞自噬水平、LC3Ⅱ/LC3I蛋白表达显著上升(P<0.05)。结论G-Rh2与DDP联合用药能够发挥协同抗肿瘤作用,其机制可能与抑制耐药蛋白表达,诱导细胞自噬相关。 Objective To explore the mechanism of ginsenoside Rh2 and the combined application of cisplatin in A549/DDP.Methods CCK-8 method was used to detect the effects of different concentrations of G-Rh2 and DDP on A549 and A549/DDP cells at different time points.The changes in cell morphology were observed by inverted microscope for 24 hours of G-Rh2 and DDP;MDC staining and immunofluorescence were used to detect autophagy;Western Blot was used to detect the expression of resistance related proteins MDR1,MRP1 and LRP,autophagy related proteins p62 and LC3.Results G-Rh2 induced growth inhibition in A549 cells and A549/DDP cells,and the inhibitory effect is related to drug concentration and action time.Compared with the control group,the cell growth was inhibited,MRP,MDR1,LRP and p62 protein expression were significantly decreased(P<0.01),while the autophagy level,LC3Ⅱ/LC3I protein expression were significantly increased(P<0.01)in the G-Rh2 group and DDP group;compared with the G-Rh2 group and DDP group,the cell growth was significantly inhibited,MRP,MDR1,LRP and p62 protein expression were significantly decreased(P<0.05),while the autophagy level,LC3Ⅱ/LC3I protein expression were significantly increased(P<0.05)in the G-Rh2+DDP group.Conclusion The combined application of G-Rh2 and DDP has inhibited the growth of A549/DDP cells,which might be related to the inhibition of drug resistance protein expression and autophagy.
