摘要
目的探讨参芪扶正注射液对人肺腺癌顺铂耐药细胞(A549/DDP)增殖、侵袭、迁移的影响及对角蛋白19(Keratin,KRT19)调控的上皮-间质转化(Epithelial-mesenchymal transition,EMT)与耐药的干预作用。方法肺腺癌顺铂耐药细胞株A549/DDP随机分为4组,对照组及药物干预组,分别给予顺铂、参芪扶正注射液及参芪扶正注射液+顺铂干预;CCK-8筛选参芪扶正注射液最佳干预浓度;Transwell检测细胞侵袭和迁移能力;EdU染色检测细胞增殖能力;Western Blot检测KRT19,EMT相关蛋白E-钙黏蛋白(E-cadherin)、波形蛋白(Vimentin)、N-钙黏蛋白(N-cadherin)及耐药相关蛋白ATP结合盒转运超家族多药耐药相关蛋白1(multidrug resistance-associated protein 1,ABCC1)、ABC蛋白G超家族成员2(ATP-binding cassette superfamily G member 2,ABCG2)的表达。结果与对照组相比,药物干预组细胞侵袭、迁移、增殖能力均显著降低,其中,参芪扶正注射液+顺铂组均较顺铂组细胞侵袭数、迁移数、增殖能力进一步下降(P<0.05);与顺铂组相比,参芪扶正注射液+顺铂组KRT19、N-cadherin、Vimentin、ABCC1、ABCG2蛋白表达显著下调(P<0.05),E-Cadherin蛋白表达显著上调,差异有统计学意义(P<0.05)。结论参芪扶正注射液可抑制A549/DDP细胞增殖、侵袭、迁移能力,提高顺铂敏感性,可能与干预KRT19调控的EMT与耐药有关。
Objective To investigate the effects of Shenqi Fuzheng Injection(参芪扶正注射液)on the proliferation,invasion and migration of cisplatin-resistant A549/DDP cells of human lung adenocarcinoma and the intervention on the epithelial-mesenchymal transition(EMT)and drug resistance regulated by Keratin(KRT19).Methods A549/DDP cells were randomly divided into 4 groups(control group and medication group).The medication groups were given cisplatin,Shenqi Fuzheng Injection and Shenqi Fuzheng Injection+cisplatin intervention respectively.CCK8 was used to selected the optimal intervention concentration of Shenqi Fuzheng Injection.Transwell was used to observe the cell invasion and migration ability.EdU staining was used to observe the cell proliferation ability.The expressions of KRT19,EMT and resistance-associated proteins(E-Cadherin,N-cadherin,Vimentin,ABCC1,ABCG2)were detected by Western Blot.Results Compared with those in the control group,the cell invasion,migration and proliferation abilities in the medication intervention group were significantly reduced.Among them,the cell invasion,migration and proliferation abilities in the Shenqi Fuzheng Injection+cisplatin group were further decreased than those in the cisplatin group(P<0.05).Compared with those of the cisplatin group,the protein expression levels of KRT19,N-cadherin,Vimentin,ABCC1 and ABCG2 in the Shenqi Fuzheng Injection+cisplatin group were significantly down-regulated(P<0.05),and E-Cadherin protein expression was significantly up-regulated(P<0.05).The differences were statistically significant.Conclusion Shenqi Fuzheng Injection can inhibit the proliferation,invasion and migration of A549/DDP cells,and increase the sensitivity of cisplatin,which may be related to the intervention of EMT and drug resistance regulated by KRT19.
