眼针抑制CIRI大鼠NF-κB信号通路活化的抗炎机制

Anti-Inflammatory Mechanism of Eye Acupuncture to Inhibit the Activation of NF-kB Signaling Pathway in CIRI Rats

摘要 目的探讨眼针对脑缺血再灌注损伤(CIRI)大鼠模型的抗炎作用机制,并通过对NF-κB信号通路关键靶点表达水平检测,阐述眼针抑制NF-κB信号通路活化的抗炎机制。方法将SPF级成年SD大鼠随机分为假手术组和造模组。采用改良线栓法建立CIRI大鼠模型,造模成功后随机分为模型对照组、眼针组和穴区外组。采用ELISA法检测脑缺血半暗带组织中IL-1β、IL-6、TNF-α,采用Western blot法检测脑组织中NF-κB、p-NF-κB、TLR4、MyD88蛋白表达水平。结果ELISA检测显示,模型对照组、眼针组及穴区外组大鼠的IL-1β、IL-6、TNF-α表达水平高于假手术组。与模型对照组对比,眼针组表达水平显著降低,穴区外组无显著差异。Western Blot法检测表明,模型对照组、眼针组和穴区外组中NF-κB、p-NF-κB、TLR4、MyD88蛋白表达水平高于假手术组。与模型对照组对比,眼针组表达水平显著降低,穴区外组无显著差异。结论眼针对CIRI模型大鼠具有抗炎作用,该作用表明是通过抑制NF-κB信号通路活化而实现的。 Objective To explore the anti-inflammatory mechanism of eye needling in CIRI rats,and to elaborate the anti-inflammatory mechanism of eye needling to inhibit the activation of NF-κB signaling pathway through the detection of the expression level of key targets of NF-κB signaling pathway.Methods SPF-grade adult SD rats were randomly divided into a sham operation group and a model group.A modified version of the thread bolus method was used to prepare the CIRI rat model,which was divided into a model control group,an eye-acupuncture group,and a group outside the acupoint area after successful modeling.The levels of IL-1β,IL-6,and TNF-α in the semi-dark zone of cerebral ischemia were detected by ELISA,and the protein expression levels of NF-κB,p-NF-κB,TLR4,and MyD88 in the brain tissues were detected by Western Blot.Results ELISA assay showed that the expression levels of IL-1β,IL-6,and TNF-α were higher in the model control group,the eye-acupuncture group,and the group outside the acupoint area than in the sham-operated group.Compared with the model control group,the expression levels were significantly lower in the eye-acupuncture group,and there was no significant dfference in the extra-cavity area group.Western Blot assay showed that the expression levels of NF-κB,p-NF-κB,TLR4,and MyD88 proteins were higher in the model control group,eye-acupuncture group and extra-cavity area group than in the sham-operated group.Compared with the model control group,the expression levels were significantly lower in the eye-acupuncture group,and there was no significant difference in the extracavity area group.Conclusion Eye acupuncture has an anti-inflammatory effect in CIRI model rats,and this effect may be achieved by inhibiting the activation of NF-κB signaling pathway.
