摘要
本文优化了刺梨多酚的溶剂提取工艺并研究其对非酒精性脂肪肝(NAFLD)的保护作用。以溶剂提取法提取刺梨多酚,通过单因素实验及四因素三水平的正交试验,考察料液比、乙醇浓度、提取次数、提取时间共4种因素对刺梨多酚提取率的影响。在高脂饮食诱导的NAFLD模型中,C57BL/6J雄性小鼠40只,随机分为正常组、模型组、高剂量刺梨组、低剂量刺梨组,连续给药4周。末次给药12 h后测定各组小鼠的体重、肝脏湿重,计算肝体指数;生化法检测血清中甘油三酯(TG)、总胆固醇(TC)、谷丙转氨酶(ALT)、草丙转氨酶(AST)的水平;油红O染色法观察肝脏的脂质沉积情况。溶剂提取刺梨多酚的最佳条件为:料液比1:15(g/mL)、乙醇浓度65%、提取次数3次、提取时间1.2 h,刺梨多酚提取率为3.11%。在此最佳提取工艺下获得的刺梨多酚提取物可有效降低NAFLD模型小鼠的体重、肝脏湿重及肝体指数(p<0.05),减少模型小鼠血清中TG、TC、ALT、AST的积累(p<0.05),同时减轻了模型小鼠肝脏的脂肪变性和脂质沉积。
Optimization of the solvent extraction process of polyphenols from Rosa roxburghi Tratt.and investigation of their protective effects against non-alcoholic fatty liver disease(NAFLD)were carried out.Polyphenols were extracted from Rosa roxburghii Tratt.by solvent extraction.Single-factor experiments and a four-factor,three-level orthogonal test were conducted to evaluate the effects of material-to-liquid ratio,ethanol concentration,extraction frequency,and extraction time on polyphenol yield.In a high-fat diet-induced NAFLD model,40 male C57BL/6J mice were randomly divided into four groups:normal control,model control,highdose polyphenol extracts,and low-dose polyphenol extracts.The mice were treated for 4 weeks.12 h After the last administration,body weight and liver wet weight were measured,and the liver index was calculated.Serum levels of triglycerides(TG),total cholesterol(TC),alanine aminotransferase(ALT),and aspartate aminotransferase(AST)were detected by biochemical assays.Hepatic lipid deposition was observed by Oil Red O staining.The optimal extraction conditions for Rosa roxburghi Tratt.polyphenols were as follows:material-to-liquid ratio of 1:15(g·mL^(-1)),65% ethanol concentration,3 extraction cycles,and 1.2 h per extraction,yielding 3.11% polyphenol.Under these conditions,the extracted polyphenols significantly reduced body weight,liver wet weight,and liver index in NAFLD model mice(p<0.05),decreased serum levels of TG,TC,ALT,and AST(p<0.05),and alleviated hepatic steatosis and lipid deposition.
