贵州省两主栽花椒品种响应干旱胁迫的转录组学分析

Transcriptomic analysis of two main Zanthoxylum bungeanum varieties response to drought stress in Guizhou Province

摘要 【目的】加速贵州省花椒栽培良种化进程,保障贵州省花椒产业健康发展。【方法】以贵州省两主栽花椒品种顶坛花椒和九叶青花椒幼苗为材料,进行水分胁迫试验,设置土壤含水量50%~75%(对照组)、30%~50%(干旱)2个处理。处理后,采集不同处理的成熟叶片进行转录组分析,获得数据进行以下处理:在数据库注释2个品种不同处理差异表达基因功能及富集通路;对2个品种不同处理叶片关键差异基因富集通路进行深入分析,探讨2个花椒品种的抗旱策略。【结果】由2个花椒品种样品共得到93.180 Gb转录数据,Q_(30)碱基占比在97%以上,GC含量为45%~48%,组装所得292192个Unigenes分别被注释到六大数据库,其中186659(33.74%)个Unigenes至少与6个数据库中的1个数据库匹配,基因注释效果较好。2个花椒品种的差异表达基因处于一个量级,九叶青花椒差异表达基因数量显著高于顶坛花椒。2个花椒品种差异表达基因KEGG注释主要富集在核糖体、碳代谢、光合作用相关途径、次级代谢物的生物合成途径、植物激素信号转导、MAPK信号途径、钙信号途径等与植物逆境响应相关的调控途径,其中顶坛花椒的响应通路更为丰富。2个花椒品种差异表达基因的相关性分析结果表明,上调基因之间的相关性高于下调基因,说明上调表达基因通路之间关系更为密切。【结论】2个花椒品种在响应干旱胁迫时具有不同的应对策略,本土品种顶坛花椒具有更加丰富的水分胁迫调控策略,引进品种九叶青花椒响应水分胁迫与光合作用途径关系密切。 【Objective】In order to accelerate the process of cultivating high-quality varieties of Zanthoxylum bungeanum in Guizhou Province and ensure the healthy development of Z.bungeanum industry in Guizhou Province.【Method】Water stress experiments were conducted on the seedlings of two main Z.bungeanum varieties of Guizhou Province,Dingtan and Jiuyeqing,with soil moisture content 50%-75%(control group)and 30%-50%(drought group).After treatment,mature leaves with different treatments were collected for transcriptome analysis to obtain data for the following processing:Annotate the differentially expressed gene functions and enrichment pathways of two varieties of Z.bungeanum with different treatments in the database.In depth analysis of key differential gene enrichment pathways in leaves of two varieties of Z.bungeanum under different treatments and exploration of drought resistance strategies for the two varieties of Z.bungeanum.【Result】A total of 93.180 Gb transcription data were obtained from 12 samples of two Z.bungeanum cultivars,with Q_(30) base percentage above 97%and GC content between 45%and 48%,annotating 292192 unigenes into six major databases,with 186659(33.74%)unigenes matching at least one of the six databases,indicating good gene annotation result.The differentially expressed genes of the two varieties of Z.bungeanum were on the same order of magnitude,and the number of differentially expressed genes in Jiuyeqing is significantly higher than that in Dingtan.The KEGG annotation of differentially expressed genes between two varieties of Z.bungeanum was mainly enriched in regulatory pathways related to plant stress response,such as ribosome,carbon metabolism,photosynthesis related pathways,biosynthesis pathways of secondary metabolites,plant hormone signal transduction,MAPK signal pathway,calcium signal pathway,etc.Among them,the response pathway of Dingtan was more abundant.Correlation analysis of differentially expressed genes between two varieties of Z.bungeanum revealed that the correlation between up-regulated genes was higher than that between down-regulated genes,indicating a closer relationship between up-regulated gene pathways.【Conclusion】The two varieties of Z.bungeanum have different coping strategies in response to drought stress.The local variety Dingtan has richer water stress regulation strategies,while the introduced variety Jiuyeqing has a close relationship with photosynthetic pathways.
