摘要
目的研究白屈菜红碱对脂多糖(LPS)诱导的类风湿关节炎(RA)成纤维样滑膜细胞MH7A细胞的MAPK/NF-κB通路表达的影响,探讨白屈菜红碱治疗RA的作用机制。方法采用LPS对MH7A细胞进行刺激,再用白屈菜红碱对刺激后的MH7A细胞进行干预,并随机分为正常组、模型组、阳性组、白屈菜红碱(高、中、低)剂量组。收集上述各组细胞,ELISA检测各组细胞上清液中TNF-α、IL-1β、IL-6的含量,RT-qPCR、Western blot检测各组细胞中ERK1/2、p38 MAPK、NF-κB p65 mRNA及蛋白的表达。结果与正常组比较,LPS处理后细胞TNF-α、IL-1β、IL-6的含量明显上升(P<0.01),ERK1/2、p38MAPK、NF-κB p65 mRNA及蛋白的表达显著升高(P<0.01);与模型组相比,阳性药物和白屈菜红碱组均改善上述指标变化。结论抑制MAPK/NF-κB通路相关因子在成纤维样滑膜细胞中的表达应是白屈菜红碱治疗RA的作用机制之一。
Objective To study the effect of chelerythrine on MAPK/NF-κB pathway expression in fibroblastic synoviocytes MH7A induced by LPS and to investigate the mechanism of action in RA.Methods MH7A cells were stimulated by LPS,and then the stimulated MH7A cells were intervened by chelerythrine that randomly divided into normal group,model group,positive group,chelerythrine high,medium and low dose groups.The cells were collected and detected by ELISA for TNF-α,IL-1βand IL-6 and expression of ERK1/2,p38 MAPK,NF-κB p65 mRNA and protein in each group by RT-qPCR and Western blot.Results Compared with the normal group,the contents of TNF-α,IL-1βand IL-6 were significantly increased after LPS treatment(P<0.01),and the mRNA and protein expressions of ERK1/2,p38 MAPK and NF-κB p65 were significantly increased(P<0.01).Compared with the model group,the positive drug and chelerythrine group could improve the above indexes.Conclusion Inhibiting the expression of MAPK/NF-κB pathway related factors in fibroblast-like synoviocytes should be one of the mechanisms of chelerythrine in the treatment of rheumatoid arthritis.
