摘要
目的分析肝细粒棘球蚴病(CE)患者术前、术后血浆外泌体微小RNA(miRNA)的表达谱。方法收集肝CE患者术前和术后及对照组(健康志愿者)血浆样本。采用纳米颗粒追踪分析、透射电子显微镜和纳米流式细胞仪对血浆外泌体的粒径、形态及表面标志物(CD9、CD63、CD81)进行表征。通过高通量测序分析血浆外泌体miRNA的表达差异,采用主成分分析和热图聚类分析评估组间miRNA表达的差异,并构建火山图直观展示差异表达miRNA。采用GO与KEGG功能富集分析差异表达miRNA的靶基因。结果术前组与术后组分离的外泌体形态完整,均呈典型的圆形或杯状结构;两组间粒径与含量差异均无显著性(P>0.05)。外泌体特异性表面标志物CD9、CD63和CD81均呈阳性表达。各组间重叠差异表达miRNA有5个。GO分析结果主要富集于对激素的反应、激酶结合、脂筏等。KEGG主要通路为癌症信号转导通路、丝裂原活化蛋白激酶信号通路和钙信号通路等。结论从肝CE患者血浆外泌体miRNA表达谱中鉴定出一系列具有成为诊断标志物及揭示疾病机制潜力的差异表达miRNA(miR-16-5p、miR-19a-3p、miR-204-3p、miR-320c、miR-582-3p)。
Aim To analyze the expression profiles of plasma exosomal microRNA(miRNA)in patients with hepatic echinococcosis(CE)before and after surgery.Methods Preoperative and postoperative plasma samples from patients with hepatic CE,as well as the control group(healthy votunteers)plasma samples were collected.The particle size distribution,morphology,and surface markers(CD9,CD63,CD81)of plasma exosomes were characterized using nanoparticle tracking analysis,transmission electron microscopy,and nanoflow cytometry,respectively.High-throughput sequencing was used to analyze the differential expression of miRNAs in plasma exosomes,principal component analysis and heatmap clustering analysis were used to evaluate the differences in expression miRNA between groups,and volcano plots were constructed to visually display differentially expressed miRNAs.GO and KEGG functional enrichment analysis were used to identify differentially expressed miRNA target genes.Results The separated exosomes in the preoperative group and in the postoperative group had a complete morphology,with typical circular or cup-shaped structures;There was no statistically significant difference in particle size and contents between the two groups(P>0.05).The exosomal specific surface markers CD9,CD63,and CD81 were all positively expressed.There are 5 overlapping differentially expressed miRNAs among the groups.The GO analysis results were mainly enriched in response to hormones,kinase binding,lipid rafts,etc.The main pathways of KEGG include cancer signaling pathway,mitogen activated protein kinase signaling pathway,and calcium signaling pathway.Conclusion A series of differentially expressed miRNAs(miR-16-5p,miR-19a-3p,miR-204-3p,miR-320c,miR-582-3p)with the potential to become diagnostic markers and reveal disease mechanisms were identified by analyzing the expression profiles of plasma exosomal miRNAs in patients with hepatic CE before and after surgery.
